3個(gè)miniSTR基因座四色熒光復(fù)合分型體系的構(gòu)建及應(yīng)用

【摘要】:目的建立D3S3053、D6S474、D20S482(擴(kuò)增片段150bp)3個(gè)miniSTR基因座四色熒光復(fù)合分型體系,用于高度降解DNA樣本的基因分型。方法采用6-FAM、HEX、TAMRA熒光染料標(biāo)記D20S482、D3S3053、D6S474基因座上游引物,構(gòu)建并優(yōu)化復(fù)合擴(kuò)增體系,在ABI 310遺傳分析儀上對(duì)擴(kuò)增產(chǎn)物進(jìn)行電泳分析,Genemapper 3.2軟件分析產(chǎn)物片段大小并進(jìn)行分型。采用上述體系對(duì)120份河北漢族健康無關(guān)個(gè)體血樣進(jìn)行檢測,并計(jì)算群體遺傳學(xué)參數(shù)。比較該體系與ID試劑盒用于高度降解檢材的成功率及靈敏度。結(jié)果D3S3053、D6S474、D20S482 3個(gè)miniSTR基因座熒光標(biāo)記復(fù)合擴(kuò)增分型體系穩(wěn)定可靠,靈敏度達(dá)50pg,用于高度降解檢材分析的成功率明顯高于ID試劑盒。3個(gè)基因座在河北漢族人群中的累積個(gè)人識(shí)別能力為0.998,累積非父排除率為0.84。結(jié)論該系統(tǒng)可用于分析高度降解DNA樣本的基因分型,進(jìn)行法醫(yī)學(xué)個(gè)人識(shí)別和親權(quán)鑒定。
[Abstract]:Objective to establish a four-color fluorescent multiplex typing system of D3S3053-D6S474D20S482 (amplified fragment 150bp) for the genotyping of highly degraded DNA samples. Methods the upstream primers of D20S482D3S3053D6S474 locus were labeled with 6-FAMHE-HEXTA-TAMRA, and the compound amplification system was constructed and optimized. ABI 310 genetic analyzer was used to analyze the amplified products by electrophoretic analysis and genotyping using Genemapper 3.2 software. The above system was used to detect the blood samples of 120 healthy unrelated individuals in Hebei Han nationality. The population genetic parameters were calculated, and the success rate and sensitivity of the system and ID kit were compared. Results the multiplex amplification system of D3S3053, D6S474, D20S482 miniSTR locus was stable and reliable. The sensitivity was 50 PG, and the success rate of high degradation detection was significantly higher than that of ID kit. The cumulative personal identification ability of three loci in Hebei Han population was 0.998, and the cumulative non-paternal exclusion rate was 0.84.Conclusion this system can be used for analysis. Genotyping of highly degraded DNA samples, To conduct forensic personal identification and paternity identification.
