藏藥鐮形棘豆醇提物酸堿分離及抗小鼠Lewis肺癌活性初步篩選
[Abstract]:Objective to screen the active parts of alcohol extract from Tibetan medicine against Lewis lung cancer-bearing mice. Methods the alcohol extract of sickle bean was separated by acid-base treatment, and the (AlF) and (AcF) were analyzed qualitatively and quantitatively. C57BL/6 mice model of Lewis lung cancer was established and randomly divided into model group, cyclophosphamide (CTX) group (20 mg kg~ (- 1), AlF high), low dose group (180, 90 mg kg~ (- 1), AcF), low dose group) (180, 90 mg kg~ (- 1),). Continuous administration for 10 days; The tumor weight, tumor inhibition rate, thymus index and spleen index of mice were measured to compare the antitumor effect of AlF,AcF, and the morphological changes of tumor cells in each group were observed under microscope by HE staining. Results AcF was mainly composed of flavonoids, the content of which was 18.9%. The content of AlF was mainly alkaloid in the high-dose 64.1%.AcF group. The tumor weight of male mice was significantly lower than that of the model group (P0.01, P0.05), and the tumor weight of male mice was significantly lower than that of the model group (P0.01, P0.05). The tumor inhibition rate was 36.3% and 32.8% respectively. Compared with the model group, the high dose of AlF could decrease the tumor weight of female mice significantly (P0.05), and the tumor inhibition rate was 29.7%. Compared with the model group, the index of spleen and thymus in CTX group decreased significantly (P 0.05, P0.01). Compared with the model group, there was no significant difference in spleen and thymus index between each dose group of AcF,AlF and the model group (P0.05). Pathological observation of tumor tissue showed different degree of necrosis of tumor cells in each group of AcF,AlF administration. Conclusion the alcohol extract AlF,AcF from sickle bean has obvious antitumor effect on Lewis lung cancer-bearing mice. The effect of flavonoids is slightly stronger than that of alkaloids, and its anti-tumor mechanism needs to be further studied.
