麝香烏龍丸對(duì)RA患者外周血miR-146a、miR-130及miR-223表達(dá)水平的影響

【摘要】:目的探討麝香烏龍丸(人工麝香、制川烏、地龍等)含藥血清對(duì)類風(fēng)濕關(guān)節(jié)炎(RA)患者外周血單個(gè)核細(xì)胞(PBMCs)中miR-146a、miR-130、miR-223表達(dá)水平的影響。方法從30例RA患者血液中提取PBMCs,將細(xì)胞分為3組,空白對(duì)照組、空白血清組、麝香烏龍丸含藥血清組。48 h后提取總RNA,采用實(shí)時(shí)熒光定量聚合酶鏈?zhǔn)椒磻?yīng)(RT-PCR)法檢測(cè)3種miR的表達(dá)水平。結(jié)果與空白血清組相比,麝香烏龍丸含藥血清組miR-146a、miR-130、miR-223表達(dá)水平顯著降低(P0.01)。結(jié)論麝香烏龍丸可通過(guò)下調(diào)PBMCs中miR-223、miR-130、miR-146a表達(dá)水平來(lái)抑制RA患者炎癥。
[Abstract]:Aim to investigate the effect of Shexiang Wulong pills (artificial musk, Radix Polygoni chuanwu, Dilong, etc.) on the expression of miR-146a,miR-130,miR-223 in peripheral blood mononuclear cells (PBMCs) of patients with rheumatoid arthritis (RA). Methods PBMCs, was extracted from the blood of 30 patients with RA and divided into three groups: blank control group, blank serum group and Shexiang Wulong pill containing serum group. 48 hours later, total RNA, was extracted. Real-time fluorescence quantitative polymerase chain reaction (RT-PCR) was used to detect the expression level of three kinds of miR. Results compared with the blank serum group, the expression level of miR-146a,miR-130,miR-223 in the Shexiang Wulong pills group was significantly lower than that in the blank serum group (P0.01). Conclusion Shexiang Wulong Pill can inhibit the inflammation of RA patients by down-regulating the expression of miR-223,miR-130,miR-146a in PBMCs.
