從RhoA通路探討電針改善腦缺血再灌注大鼠神經(jīng)功能的機(jī)制
[Abstract]:[objective] to investigate the effects of electroacupuncture on the neural function of middle cerebral artery occlusion (MCAO) rats with (MCAO) ischemia-reperfusion (I / R) and its potential mechanism. [methods] 54 male SPF grade SD rats were randomly divided into sham operation group, cerebral infarction group and electroacupuncture group with 18 rats in each group according to the method of random numerical table, and 54 male SD rats with acute cerebral infarction were divided into sham operation group, cerebral infarction group and electro acupuncture group with 18 rats in each group. Sham operation group only do operation trauma, cerebral infarction group without any intervention, electroacupuncture group to acupuncture Baihui point, Dazhui point, once a day, 30 min. each time. At each time point (1,7,14 days), the neurologic function was assessed by the nerve function defect scale, and the A (RhoA), (A (RhoA), a member of the Nogo-A), Ras homologue gene family) was detected by protein immunoblotting (Western blot) in the focal side of the hippocampus. [results] the expression of lysophosphatidic acid (LPA) protein in cerebral infarction group was significantly higher than that in sham operation group on day 1,7,14 (P0.05), and the expression of Nogo-A and LPA protein in hippocampus of cerebral infarction group was significantly higher than that in sham operation group (P0.05). On the 7th and 14th day, the expression of RhoA protein in cerebral infarction group was significantly higher than that in sham operation group (P0.05). On the 1st, 7th and 14th day, the nerve function defect score, the expression of RhoA,Nogo-A and LPA protein in the hippocampus of the acupuncture group were significantly lower than those in the cerebral infarction group at the same time point (P0.05). [conclusion] electroacupuncture can improve the nerve function after cerebral infarction in rats. The mechanism may be related to the inhibitory effect of electroacupuncture on the expression of RhoA pathway-related proteins in hippocampus of rats.
