“秩邊透水道”針?lè)ㄖ委煷笫笤l(fā)性痛經(jīng)作用機(jī)制的實(shí)驗(yàn)研究
[Abstract]:Objective to observe the general behavior, writhing times, serum interleukin-1 (IL-1) and interleukin-6 (IL-6) in rats with dysmenorrhea by establishing a rat model of primary dysmenorrhea. Tumor necrosis factor-a (TNF-a), plasma 尾 -endorphin (尾 -endorphin) and 5-hydroxytryptamine (5-HT) in hypothalamus were analyzed. To provide accurate experimental basis for the emergence of new clinical technology, in order to better promote clinical. Methods Forty healthy female SD rats (230g 鹵15g) were randomly divided into four groups: blank group, model group, acupuncture group and rank side group. The model of dysmenorrhea induced by oxytocin: the rats in the model group, acupuncture group and rank side group were injected estradiol benzoate subcutaneously into the femoris for 10 days, only 0.5mg/ on the 1st day, 0.2mg/ on the 2nd to 9th day, and 1 hour after the last administration of estradiol benzoate. Intraperitoneal injection of oxytocin 2u/; Rats in the blank group were subcutaneously injected with the same dose of normal saline daily. Intervention was carried out from the first day of modeling for 5 days each time for 20 minutes. The blank group and the model group only grasped fixation. The acupuncture group needled Guanyuan after modeling every day, and the acupoints of Sanyinjiao on both sides were punctured. In the rank side group, the needle intervention of "rank edge permeating waterway" was carried out immediately after making model every day. In the course of modeling and treatment, we observed the general condition of color, diet, exercise and body weight of rats, recorded the times of writhing in 30 minutes after intraperitoneal injection of oxytocin, and detected the serum IL-1,IL-6, of rats by radioimmunoassay. The content of 5-HT in hypothalamus tissue of plasma 尾-EP, in TNF-a, was analyzed statistically and the results were summarized. Results 1. The normal condition of the blank group was good and no obvious abnormality was found. In the model group, the hair color was dim and yellowish, especially in the abdomen with obvious yellowish humidity, decreased appetite, slow movement and weight loss. Compared with the model group, the acupuncture group and the rank side group showed significant improvement. 2. There was no writhing reaction in the blank group, all the rats in the model group, the acupuncture group and the rank side group had writhing reaction, and the writhing reaction was more obvious in the model group. The contents of plasma 尾-EP in the model group were higher than those in the blank group, the contents of 尾-EP in the rank side group and acupuncture group were higher than those in the model group, and the rank side group was higher than that in the acupuncture group, and the content of 尾-EP in the rank side group was higher than that in the acupuncture group. The difference was significant (P0.05). Compared with the blank group the content of 5-HT in the hypothalamus of the model group was significantly lower than that of the blank group (P0.05). In acupuncture group, rank side group and model group, the content of 5-HT in hypothalamus was significantly different (P0.05), but there was no significant difference between rank side group and acupuncture group. Compared with the blank group, the serum TNF-a,IL-1,IL-6 content in the model group was significantly higher than that in the blank group (P0.05). Compared with the model group, the serum TNF-a,IL-1,IL-6 content in acupuncture group, rank edge group and model group was significantly different (P0.05), but there was no significant difference between the rank side group and acupuncture group. Conclusion the experimental model of primary dysmenorrhea in rats is successful. The acupuncture method of "rank side through waterway" could obviously improve the general condition of primary dysmenorrhea rat model, decrease the number of writhing, increase the content of 5-HT and plasma 尾-EP in hypothalamus, and decrease the content of serum IL-1,IL-6,TNF-a. The mechanism of treatment of primary dysmenorrhea is elucidated preliminarily, which is beneficial to the connection between basic research and clinical application, and provides experimental support for the emergence of new technology.
