補(bǔ)腎活血方對(duì)URSA患者血清中IL-13、IL-15、IL-35的影響研究
[Abstract]:Objective: To study the regulation mechanism and clinical effect of Bushen Huoxue recipe for unexplained recurrent spontaneous abortion (URSA), and to select the three factors of interleukin-13 (IL-13), interleukin-15 (IL-15) and interleukin-35 (IL-35). The expression of IL-13R-1, IL-15R-1, IL-12R-2 and gp130 in the decidua tissues of the patients with URSA and normal early pregnancy were observed by observing the expression of IL-13R-1, IL-15R-1, IL-12R-2 and gp130 in the decidua tissues of the patients with URSA and normal early pregnancy. To study the effects of Bushen Huoxue Recipe on IL-13, IL-15 and IL-35, the relationship between IL-13, IL-15 and IL-35 and URSA was explored. Methods: The experimental study was divided into two parts. The first part collected 15 cases of non-unexplained recurrent spontaneous abortion with unexplained recurrent spontaneous abortion during the period from January to March 2017 in the Affiliated Hospital of Heilongjiang University of Traditional Medicine, Obstetrics and Gynecology and in the hospital for 6-10 weeks as the study group (URSA group). In the second part, 60 patients with URSA were collected, and the peripheral blood was collected and divided into 30 cases, the experimental group with the Bushen Huoxue and the dydrogesterone group, and the control group of the dydrogesterone in 30 cases. The peripheral blood was collected before and after the administration of the two groups. The expression of IL-13, IL-15 and IL-35 in 15 patients with URSA and 15 normal early pregnancy women were detected by RT-PCR, and the levels of IL-13, IL-15 and IL-35 in peripheral blood were determined by ELISA. The level of estrogen and HCO3-HCG was changed. Results: (1) The expression of IL-13R-1 and gp130 in decidua tissues of URSA was significantly lower than that of normal people, while the expression of IL-15R-1 and IL-12R-1 was significantly higher than that of normal control group (P <0.05). (2) The content of IL-13 in the peripheral blood of the URSA experimental group was significantly higher than that of the control group. The content of IL-13 in the peripheral blood of the control group was significantly higher than that of the control group. The content of IL-13 in the peripheral blood of the control group was higher than that of the control group (P0.01). (3) The content of IL-15 in the peripheral blood of the patients with URSA was significantly lower than that in the control group. The content of IL-15 in the peripheral blood of the control group was significantly lower than that of the control group. The content of IL-15 in the peripheral blood of the control group was lower than that of the control group (P0.01). (4) The content of IL-35 in the peripheral blood of the URSA experimental group was significantly higher than that of the control group, and the level of IL-35 in the peripheral blood of the control group was significantly higher than that of the control group (P0.01). (5) The content of progesterone, estradiol and blood-HCG in the peripheral blood of the experimental group of the Bushen Huoxue and the dydrogesterone group was significantly higher than that before treatment (P0.01). Conclusion: (1) The expression of IL-13R-1 and gp130 in the decidua tissue of the patients with URSA is decreased compared with that of the normal pregnancy and the expression of IL-15R-1 and IL-12R-2 is increased, indicating that the IL-13, IL-15 and IL-35 cytokines play a role in the decidua tissues of the patients with URSA and the normal early pregnancy, and it is suggested that IL-13 and IL-15, IL-35 may be involved in the pathogenesis of the URSA. (2) The content of IL-13 and IL-35 in the peripheral blood of both the URSA and the control group was significantly higher than that of the control group. The content of IL-15 in the peripheral blood of the URSA experimental group and the control group was significantly lower than that in the control group, indicating that the appropriate amount of IL-15 was beneficial to the maintenance of pregnancy, and the high concentration of IL-15 was not conducive to the pregnancy, which may be one of the causes of the abortion. (3) Bushen Huoxue Recipe may improve the level of IL-13 and IL-35 in the peripheral blood of the URSA experimental group, and decrease the level of the factor of IL-15 in the peripheral blood of the URSA experimental group to improve the clinical curative effect of the patients with recurrent spontaneous abortion. (4) Bushen Huoxue Decoction can obviously improve the content of progesterone, estradiol and blood-HCG in the peripheral blood of the patients with URSA, so as to maintain the pregnancy.
