溫陽(yáng)益氣方對(duì)潰瘍性結(jié)腸炎大鼠Treg細(xì)胞的免疫調(diào)節(jié)作用

【摘要】:目的:觀(guān)察溫陽(yáng)益氣方對(duì)由三硝基苯磺酸(TNBS)/乙醇誘導(dǎo)的潰瘍性結(jié)腸炎(UC)大鼠模型Treg細(xì)胞的影響,進(jìn)而探討溫陽(yáng)益氣方治療UC的作用機(jī)制和靶點(diǎn)。方法:經(jīng)過(guò)TNBS/乙醇造模成功后,分為空白組、溫陽(yáng)益氣(WYYQ)組、模型組、美沙拉嗪(MT)組。用灌胃的方式給藥持續(xù)14天后,把大鼠處死并取標(biāo)本。觀(guān)察UC大鼠結(jié)腸粘膜組織的病理改變采用蘇木精-伊紅染色(HE)法。檢測(cè)大鼠血清中IL-10的表達(dá)用酶聯(lián)免疫吸附試驗(yàn)(ELISA);檢測(cè)特異性轉(zhuǎn)錄因子Foxp3mRNA的基因水平用實(shí)時(shí)熒光定量PCR法。結(jié)果:1.與空白組相比,模型組大鼠出現(xiàn)神態(tài)萎靡、反應(yīng)遲鈍、弓背蜷縮、毛質(zhì)稀松、活動(dòng)極少、體溫降低、便溏、進(jìn)食不佳、體重減輕等一系列類(lèi)似陽(yáng)虛的表現(xiàn)。2.HE染色后電鏡下病理切片結(jié)果示:與空白組相比,模型組大鼠的結(jié)腸粘膜水腫、充血、糜爛明顯,腺體排列紊亂甚至斷裂消失,炎細(xì)胞(中性粒細(xì)胞、淋巴細(xì)胞)浸潤(rùn)嚴(yán)重,粘膜上皮組織嚴(yán)重?fù)p傷,多處隱窩潰瘍形成。病理組織評(píng)分模型組與空白組相比分值顯著增高,且差異有統(tǒng)計(jì)學(xué)意義(P0.05);MT組和WYYQ組的分值與模型組相比顯著降低,且差異有統(tǒng)計(jì)學(xué)意義(P0.05)。3.與空白組相比,大鼠血清中IL-10含量在模型組中濃度顯著降低,且差異有統(tǒng)計(jì)學(xué)意義(P0.05);MT組和WYYQ組與模型組相比較,大鼠血清中IL-10的濃度含量顯著增高,且差異有統(tǒng)計(jì)學(xué)意義(P0.05);MT組大鼠血清中IL-10的濃度含量較WYYQ組低,且差異有統(tǒng)計(jì)學(xué)意義(P0.05)。4.與空白組相比,模型組中Foxp3mRNA在大鼠結(jié)腸組織中的含量表達(dá)顯著減少,且差異有統(tǒng)計(jì)學(xué)意義(P0.05);MT組和WYYQ組結(jié)腸組織中Foxp3 mRNA含量表達(dá)與模型組相比,水平顯著增高,且差異有統(tǒng)計(jì)學(xué)意義(P0.05)結(jié)論:溫陽(yáng)益氣方可通過(guò)增加血清中IL-10的含量,上調(diào)結(jié)腸組織中Foxp3的表達(dá),增加機(jī)體Treg細(xì)胞含量,提高機(jī)體免疫力,從而有利于治療UC。
[Abstract]:Aim: to observe the effect of Wenyang Yiqi recipe on Treg cells of (UC) rat model of ulcerative colitis induced by trinitrobenzene sulfonic acid (TNBS) / ethanol, and to explore the mechanism and target of WYYQF in the treatment of UC. Methods: after TNBS/ ethanol was successfully established, the rats were divided into three groups: blank group, (WYYQ) group, model group and (MT) group. Rats were killed and specimens were taken after 14 days of gavage. The pathological changes of colonic mucosa in UC rats were observed by hematoxylin-eosin staining (HE) method. The expression of IL-10 in rat serum was assayed by enzyme linked immunosorbent assay (ELISA);) to detect the gene level of specific transcription factor (Foxp3mRNA) by real-time fluorescence quantitative PCR method. Results: 1. Compared with the blank group, the rats in the model group appeared to be sluggish, slow to react, curled up on the back of the bow, fluffy hair, very little activity, hypothermia, loose stools, and poor food intake. 2.HE staining showed that compared with the blank group, the colonic mucosal edema, hyperemia, erosion, gland arrangement disorder and even rupture disappeared in the model group. Inflammatory cells (neutrophils, lymphocytes) infiltration, mucosal epithelial tissue damage, many recess ulcer formation. The pathological tissue score in the model group was significantly higher than that in the blank group, and the difference was statistically significant (P0.05). (P0.05) the score of); MT and WYYQ group was significantly lower than that of the model group, and the difference was statistically significant (P0.05). Compared with the blank group, the concentration of serum IL-10 in the model group decreased significantly, and the difference was statistically significant (P0.05). Compared with model group, the concentration of IL-10 in serum of MT group and WYYQ group was significantly higher than that of model group (P0.05). The concentration of IL-10 in serum of MT group was lower than that of WYYQ group, and the difference was statistically significant (P0.05). Compared with the blank group, the expression of Foxp3mRNA in the colonic tissue of the model group was significantly decreased, and the difference was statistically significant (P0.05). The expression of Foxp3 mRNA in colon tissue of MT group and WYYQ group was significantly higher than that of model group, and the difference was statistically significant (P0.05) conclusion: Wenyang Yiqi decoction could increase the content of IL-10 in serum. The expression of Foxp3 in colon tissue is up-regulated, the content of Treg cells is increased, and the immunity of the body is enhanced, which is beneficial to the treatment of UC..
