電針對功能性消化不良大鼠GAS和SS的影響

【摘要】:目的以功能性消化不良(FD)大鼠為研究對象,觀察電針對功能性消化不良模型大鼠一般情況,檢測尿D-木糖排泄率、胃蛋白酶含量及胃排空率和小腸推進率,探討脾虛氣滯證候模型大鼠的制備,及電針該模型大鼠胃腸消化功能的影響;檢測血清和胃組織中胃泌素(GAS)、生長抑素(SS)的表達,探討GAS和SS在電針調(diào)節(jié)胃腸功能中發(fā)揮的作用。進一步明確電針治療FD的作用機理。方法選取24只雄性SD大鼠,按照隨機數(shù)字表法分為空白組、模型組、電針組,每組8只。除空白組外,其余各組均采用夾尾刺激法配合隔日進食制備FD大鼠模型,造模成功后,電針組給予電針“足三里”及“太沖”治療,空白組、模型組予束縛固定,每日1次,連續(xù)治療14d。實驗過程中觀測各組大鼠一般情況和尿D-木糖排泄率,治療結(jié)束后,檢測胃排空率,小腸推進率及胃蛋白酶含量,蘇木素-伊紅(HE)染色觀察胃竇組織,雙抗體夾心酶聯(lián)免疫吸附測定法(ELISA)測定血清和胃竇中GAS、SS的表達水平。結(jié)果1.一般情況觀察比較,造模后,與空白組相比,模型組、電針組可見大鼠煩躁、飲水、飲食減少、毛發(fā)枯黃無光澤、大便稀溏,兩組鼠體重明顯輕于空白組,差異具有統(tǒng)計學(xué)意義(p0.05)。電針組經(jīng)治療后,一般情況較模型組有所改善,大鼠體重較模型組增長明顯,差異具有統(tǒng)計學(xué)意義(p0.05),且與空白組差異無統(tǒng)計學(xué)意義(p0.05)。2.胃排空率比較,與空白組相比,模型組大鼠的胃排空率降低,差異具顯著性差異(p0.01);與模型組相比,電針組大鼠胃排空率增加,差異具有統(tǒng)計學(xué)意義(p0.05)。3.小腸推進率比較,與空白組相比,模型組大鼠的小腸推進率顯著下降,差異具有統(tǒng)計學(xué)意義(p0.01);與模型組相比,電針組大鼠的小腸推進率升高,差異具有統(tǒng)計學(xué)意義(p0.05)。4.胃蛋白酶含量比較,與空白組相比,模型組大鼠的胃蛋白酶含量減少,差異具有統(tǒng)計學(xué)意義(p0.01);與模型組相比,電針組大鼠的胃蛋白酶含量增加,差異具有統(tǒng)計學(xué)意義(p0.05)。5.尿D-木糖排泄率比較,與空白組相比,模型組大鼠的尿d-木糖排泄率降低,差異具有統(tǒng)計學(xué)意義(p0.05);與模型組相比,電針組大鼠的尿d-木糖排泄率升高,差異具有統(tǒng)計學(xué)意義(p0.05)。6.胃竇和血清中GAS和SS含量比較,與空白組相比,模型組大鼠胃竇和血清中g(shù)as含量顯著降低,ss含量顯著升高,差異均具有統(tǒng)計學(xué)意義(p0.01);與模型組相比,電針組大鼠胃竇和血清中g(shù)as含量升高,差異具有統(tǒng)計學(xué)意義(p0.05),ss含量明顯降低,差異具有統(tǒng)計學(xué)意義(p0.01)。結(jié)論1.通過夾尾刺激+隔日進食的方法造模后,大鼠食量減少、體重下降,胃竇解剖肉眼未見器質(zhì)性病變,說明造模成功。2.電針組體重、胃排空率、小腸推進率、胃蛋白酶含量、尿D-木糖排泄率都增加,說明電針具有治療作用,可以改善胃腸道功能。3.電針組胃竇和血清中GAS和SS含量變化,與胃排空率、小腸推進率、胃蛋白酶含量、尿D-木糖排泄率的相關(guān)性。電針對GAS、SS表達的調(diào)節(jié)作用可能是電針治療FD的機制之一。
