耳穴經(jīng)皮電刺激對顳葉癲癇大鼠癲癇發(fā)作頻率與海馬區(qū)膠質(zhì)細(xì)胞活性及炎性因子的影響

【摘要】:目的:觀察經(jīng)皮電刺激耳迷走神經(jīng)支配的耳穴對顳葉癲癇大鼠自發(fā)癲癇發(fā)作頻率、海馬區(qū)膠質(zhì)細(xì)胞活性及促炎細(xì)胞因子白細(xì)胞介素(IL)-6、抗炎細(xì)胞因子IL-10表達(dá)的影響,探討耳穴刺激對顳葉癲癇的緩解效應(yīng)及其可能的細(xì)胞與分子機(jī)制。方法:SD大鼠隨機(jī)分為正常組、模型組、治療組,每組12只。采用氯化鋰-匹羅卡品多次小劑量重復(fù)注射制備慢性顳葉癲癇大鼠模型。對治療組耳甲腔內(nèi)耳穴"心""肺""皮質(zhì)下"進(jìn)行經(jīng)皮電刺激干預(yù),每日1次,每次20min,治療6周。用長時程視頻系統(tǒng)監(jiān)測大鼠的癲癇發(fā)作情況,治療后用免疫熒光與Real-time PCR方法觀察癲癇大鼠海馬區(qū)小膠質(zhì)細(xì)胞標(biāo)記物Iba 1、星形膠質(zhì)細(xì)胞標(biāo)記物膠質(zhì)纖維酸性蛋白(GFAP)及IL-6、IL-10的表達(dá)情況。結(jié)果:模型組大鼠癲癇自發(fā)頻率隨時間延長顯著增加,治療組大鼠的自發(fā)癲癇頻率較模型組顯著降低(P0.05)。與正常組比較,模型組大鼠海馬CA 1、CA 3區(qū)Iba 1、GFAP、IL-6免疫陽性細(xì)胞數(shù)明顯增多(P0.05),IL-6mRNA表達(dá)水平顯著上調(diào)(P0.05)。與模型組比較,治療組大鼠海馬CA 1、CA 3區(qū)Iba 1、GFAP、IL-6免疫陽性細(xì)胞數(shù)明顯減少(P0.05),IL-6mRNA表達(dá)水平顯著下調(diào)(P0.05),IL-10陽性細(xì)胞數(shù)及mRNA表達(dá)水平顯著上調(diào)(P0.05)。結(jié)論:耳迷走神經(jīng)支配的耳穴經(jīng)皮電刺激干預(yù)能顯著降低慢性顳葉癲癇大鼠的癲癇發(fā)作頻率,此效應(yīng)可能與下調(diào)海馬區(qū)膠質(zhì)細(xì)胞活性,下調(diào)海馬內(nèi)促炎細(xì)胞因子IL-6,上調(diào)抗炎細(xì)胞因子IL-10的蛋白及基因表達(dá)水平密切相關(guān)。
[Abstract]:Objective: to observe the effects of transcutaneous stimulation of auricular points innervated by auricular vagus on the frequency of spontaneous seizure, glial cell activity in hippocampus and the expression of interleukin-6, anti-inflammatory cytokine IL-10 in rats with temporal lobe epilepsy. To investigate the effect of auricular point stimulation on temporal lobe epilepsy and its possible cellular and molecular mechanisms. Methods: Sprague-Dawley rats were randomly divided into normal group, model group and treatment group with 12 rats in each group. The rat model of chronic temporal lobe epilepsy was established by repeated injection of lithium chloride and pilocarpine. The treatment group was treated with transcutaneous electrical stimulation of heart, lung and subcortical acupoints, once a day for 20 min, for 6 weeks. Long-term video system was used to monitor epileptic seizures in rats. The expressions of microglial markers Iba1, glial fibrillary acidic protein (GFAPs) and IL-6 IL-10 in hippocampal area of epileptic rats were observed by immunofluorescence and Real-time PCR after treatment. Results: the spontaneous frequency of epilepsy in the model group was significantly increased with the prolongation of time, and the frequency of spontaneous epilepsy in the treatment group was significantly lower than that in the model group. Compared with the normal group, the number of IL-6 immunoreactive cells in the CA1 / CA3 region of the model group was significantly increased. Compared with the model group, the number of IL-6 immunopositive cells in the CA1 / CA3 region of the hippocampus of the treatment group was significantly decreased. Conclusion: transcutaneous electrical stimulation of auricular points innervated by auricular vagus nerve can significantly decrease the frequency of epileptic seizures in rats with chronic temporal lobe epilepsy. This effect may be related to the down-regulation of glial cell activity in hippocampus. Down-regulation of IL-6 and up-regulation of protein and gene expression of anti-inflammatory cytokine IL-10 are closely related.
