【摘要】：目的:1)以p38MAPK信號(hào)轉(zhuǎn)導(dǎo)通路作為切入點(diǎn),探討破裂型腰椎間盤突出模型大鼠基于p38MAPK的磷酸化與靶標(biāo)的腰間盤組織中表達(dá)MMP-7和MMP-3蛋白相互影響的關(guān)系。2)觀察益氣活血方對(duì)裂型腰椎間盤突出大鼠椎間盤組織p38MAPK信號(hào)轉(zhuǎn)導(dǎo)通路的影響,初步探討其改善的分子機(jī)制。方法:建立腰椎間盤突出重吸收大鼠模型,大鼠隨機(jī)分為模型組、益氣活血方組、益氣活血方+P38抑制劑組和P38抑制劑組,另設(shè)假手術(shù)組(n=10),假手術(shù)組及模型組灌胃純凈水,其他各組大鼠分別灌胃相應(yīng)的藥物,干預(yù)3周,3周后麻醉處死大鼠,采用雙抗體夾心法(ELISA)、HE和Western-Blot法測(cè)定血清中TXB_2和PGF_(1α)的含量和椎間盤組織病理學(xué)改變,以及椎間盤組織中P-p38MAPK及MMP-3、MMP-7蛋白表達(dá)。結(jié)果:1)模型組大鼠腰間盤組織可見有間質(zhì)輕度水腫,并有少許血管增多,單核炎細(xì)胞浸潤(rùn)及少量纖維組織增生,部分可見有神經(jīng)萎縮。給予益氣活血方治療后,其病程均有所改善。2)益氣活血方(“益氣活血”法代表方)干預(yù)破裂型腰椎間盤突出模型大鼠后,結(jié)果顯示血清中TXB_2含量明顯下降,PGF_(1α)含量有所升高,而TXB_2/PGF_(1α)比值明顯下降,均向良性轉(zhuǎn)化的趨勢(shì);與模型組相比較(P值均0.01),結(jié)果具有顯著性差異。3)與模型組比較,益氣活血方組P-p38MAPK和MMP-3、MMP-7蛋白表達(dá)均增高,結(jié)果具有顯著性差異(P0.05或P0.01);與抑制劑組比較,益氣活血方+抑制劑組P-p38MAPK和MMP-3、MMP-7蛋白表達(dá)均增高,結(jié)果具有顯著性差異(P0.05或P0.01)。結(jié)論:益氣活血方治療后病變程度有所改善,其作用機(jī)制可能激活p38MAPK信號(hào)通路而提高椎間盤組織MMP-3、MMP-7蛋白表達(dá)有關(guān)。
[Abstract]:Objective: to use p38 MAPK signal transduction pathway as the entry point. To investigate the relationship between phosphorylation of p38 MAPK and the expression of MMP-7 and MMP-3 protein in the target lumbar disc of rat model of ruptured lumbar intervertebral disc herniation. 2) to observe the effect of Yiqi Huoxue recipe on intervertebral disc tissue of rat with cleft lumbar intervertebral disc herniation. The effect of p38 MAPK signal transduction pathway, The molecular mechanism of its improvement was preliminarily discussed. Methods: a rat model of reabsorption of lumbar disc herniation was established. Rats were randomly divided into three groups: model group, P38 inhibitor group and P38 inhibitor group. Rats in other groups were administrated with corresponding drugs respectively. After 3 weeks and 3 weeks of intervention, the rats were killed under anesthesia. The contents of TXBStack2 and PGFS-1 偽 in serum and histopathological changes of intervertebral disc were measured by double antibody sandwich method and Western-Blot method. And the expression of P-p38 MAPK and MMP-3-7 protein in intervertebral disc tissue. Results in the model group, there were slight edema in the lumbar intervertebral disc, a little increase in blood vessels, infiltration of mononuclear cells and hyperplasia of a few fibrous tissues, and some neuroatrophy was found in some of the rats in the model group. After the treatment of Yiqi Huoxue recipe, the course of disease was improved. 2) after the intervention of Yiqi Huoxue recipe ("Yiqi Huoxue" method), the results showed that the content of TXBS-2 in serum decreased obviously and the content of PGFS-1 偽 increased. Compared with the model group, the ratio of TXB _ 2 / PGF _ (1 偽) was significantly decreased, and the expression of P-p38 MAPK and MMP-3 / MMP-7 in the Yiqi Huoxue recipe group was significantly higher than that in the model group, and the P value was 0.01 in comparison with the model group, and the expression of MMP-3 and MMP-3 was significantly higher in the Yiqi Huoxue recipe group than in the model group. Results compared with the inhibitor group, the expression of P-p38 MAPK and MMP-3 MMP 7 in the Yiqi Huoxue prescription inhibitor group was significantly higher than that in the control group. The results showed that there was significant difference between the two groups (P0.05 or P0.01). Conclusion: the pathological degree of Yiqi Huoxue prescription is improved after treatment, and its mechanism may be related to the activation of p38 MAPK signaling pathway and the increase of MMP-3 and MMP-7 protein expression in intervertebral disc tissue.
