【摘要】：目的:探討"長強"穴藥線植入對肛門部切口痛模型大鼠止痛的作用機制。方法:SD大鼠隨機分為空白組、模型組、長強穴藥線植入組、非經(jīng)非穴藥線植入組,每組10只。建立肛門部切口痛模型,長強穴藥線植入組用藥線植入"長強"穴,非經(jīng)非穴藥線植入組用藥線植入脅下非經(jīng)非穴點。各組于術前及術后4、8、12、24h用Von Frey纖維絲測定大鼠術區(qū)及肛門左側距術區(qū)15 mm處傷害性機械刺激的回縮閾值(MWT)。采用免疫組化SP法檢測脊髓磷酸化p 38絲裂原活化蛋白激酶(p-p 38MAPK)水平。結果:與空白組比較,模型組術區(qū)及距術區(qū)15mm處MWT在術后4、8、12、24h均明顯降低(P0.05),脊髓p-p 38MAPK水平明顯升高(P0.05)。與模型組比較,長強穴藥線植入組術區(qū)MWT在術后8、12、24h明顯升高(P0.05),距術區(qū)15mm處MWT在術后12、24h明顯升高(P0.05),術后24h脊髓p-p 38 MAPK水平明顯降低(P0.05)。非經(jīng)非穴藥線植入組術區(qū)及距術區(qū)15 mm處MWT、術后24h脊髓p-p 38MAPK水平與模型組比較,差異均無統(tǒng)計學意義(P0.05)。結論:"長強"穴藥線植入能顯著提高肛門部切口痛模型大鼠的機械痛閾,其機制可能與"長強"藥線植入降低切口痛大鼠脊髓p-p 38 MAPK水平有關,非經(jīng)非穴藥線植入不具有相同作用。
[Abstract]:Objective: to investigate the mechanism of analgesic effect of "Changqiang" acupoint line implantation on anus incision pain model rats. Methods Sprague-Dawley rats were randomly divided into blank group, model group, Changqiang acupoint line implantation group and non-meridian non-acupoint drug line implantation group with 10 rats in each group. The anus incision pain model was established. The "Changqiang" point was implanted into the drug line of Changqiang acupoint implantation group, and the non-meridian non-point was implanted under the hypochondria in the non-meridian drug line implantation group. Von Frey filament was used to determine the retraction threshold of nociceptive mechanical stimulation at 15 mm from the operation area and the left anal area in each group. The level of p38 mitogen-activated protein kinase (pp38 MAPK) in spinal cord was detected by immunohistochemical SP method. Results: compared with the blank group, the level of p-p38 MAPK in the spinal cord increased significantly in the model group and in the 15mm area after operation. Compared with the model group, the level of p-p38 MAPK in the spinal cord decreased significantly at 24 hours after operation in the Changqiang acupoint line implantation group, and at 24 hours after the operation, the level of p-p38 MAPK in the spinal cord decreased significantly at 24 hours after the operation, and at 24 hours after the operation, the level of p-p38 MAPK in the spinal cord decreased significantly. There was no significant difference in the level of p-p38 MAPK between the two groups 24 hours after operation compared with the model group. There was no significant difference in the level of p-p38 MAPK between the two groups (P 0.05). Conclusion: "Changqiang" drug line implantation can significantly increase the mechanical pain threshold of rats with anal incision pain, and the mechanism may be related to the decrease of p-p38 MAPK level in spinal cord of "Changqiang" group. Non-meridian non-acupoint line implantation does not have the same effect.
