壯藥金鈕扣質(zhì)量標(biāo)準(zhǔn)研究

【摘要】:目的:對壯藥金鈕扣質(zhì)量標(biāo)準(zhǔn)進(jìn)行研究,并建立其高效液相色譜指紋圖譜的方法。方法:采用四大鑒別法,即原植物鑒別法、藥材性狀鑒別法、顯微鑒別法以及理化鑒別法,對該藥進(jìn)行生藥學(xué)鑒別的研究;按照《中華人民共和國藥典》2015年版四部通則中的方法對該藥的水分、灰分等常規(guī)限量檢查以及浸出物含量進(jìn)行研究;依據(jù)化學(xué)成分的定性反應(yīng),對該藥的化學(xué)成分進(jìn)行研究;運(yùn)用高效液相色譜法對壯藥金鈕扣藥材中綠原酸成分進(jìn)行含量測定以及建立該藥的指紋圖譜。結(jié)果:(1)金鈕扣植物的莖呈綠色,并帶有紫紅色,有縱向條紋,葉為對生,葉片呈寬卵圓形或橢圓形,頂端短尖或稍鈍,全緣或?yàn)椴钼g鋸齒,花黃色,為頭狀花序;藥材性狀特征為莖呈黃綠色,有縱條紋,斷面類白色,中空,葉紙質(zhì),皺縮,花灰黃色,頂生或腋生;根橫切面中皮層可見25~35個通氣組織環(huán)列,內(nèi)皮層明顯;莖橫切面中,可見非腺毛,有10~15個外韌型維管束環(huán)列,髓部寬廣,常中空;葉橫切面中,表皮可見非腺毛及氣孔,主脈維管束常3束;氣孔軸式為不定式;花粉粒、非腺毛、木栓組織等為該藥的全草粉末特征;薄層色譜鑒別中,金鈕扣供試品的薄層色譜與對照藥材和對照品色譜相應(yīng)的位置上,顯相同的藍(lán)色的熒光斑點(diǎn)。(2)初步擬定,金鈕扣藥材水分含量、總灰分含量及酸不溶性灰分含量不得高于15.0%、23.0%以及7.0%,水溶性浸出物的含量不得少于18.0%。(3)金鈕扣中可能含有有機(jī)酸、黃酮類、植物甾醇、香豆素、三萜、內(nèi)酯、酚類、揮發(fā)油等化學(xué)成分類型;金鈕扣中綠原酸含量在進(jìn)樣量為0.0100~0.4010μg范圍內(nèi)與峰面積呈良好線性關(guān)系,回歸方程為y=2643.72866x-11.165153(r=0.9997),平均加樣回收率為102.82%(RSD=1.90%),各批次金鈕扣綠原酸含量在0.0043%~0.0857%之間。(4)初步建立起壯藥金鈕扣藥材的高效液相色譜指紋圖譜,篩選出了6個共有峰。結(jié)論:通過生藥學(xué)鑒別研究所得出的金鈕扣的特征,穩(wěn)定、可靠,可以作為金鈕扣定性鑒別的依據(jù)。金鈕扣藥材在加工時,應(yīng)注意清潔,以免帶入過多的泥沙,并充分干燥,防止發(fā)霉變質(zhì),以利于貯藏。采用高效液相色譜法對金鈕扣中綠原酸含量進(jìn)行測定,該方法準(zhǔn)確、簡便、快速,重復(fù)性好。本研究為建立壯藥金鈕扣藥材的質(zhì)量標(biāo)準(zhǔn)奠定了實(shí)驗(yàn)基礎(chǔ),為其今后的合理開發(fā)及利用提供了科學(xué)的參考依據(jù)。
[Abstract]:Objective: to study the quality standard of Zhuang Yao Jin Niu and to establish a high performance liquid chromatographic fingerprint. Methods: the pharmacokinetic identification of the drug was studied by four methods, I. e., the original plant identification method, the medicinal material character identification method, the microscopic identification method and the physicochemical identification method. According to the methods in the four general principles of the 2015 edition of the Pharmacopoeia of the people's Republic of China, the water, ash and extract contents of the drug were studied, and the chemical constituents of the drug were studied according to the qualitative reaction of the chemical constituents. High performance liquid chromatography (HPLC) was used to determine the content of Lv Yuan acid in Jinniu medicinal material and to establish its fingerprint. Results the stems of the golden button plant were green and purplish red, with longitudinally striate stripes, the leaves were opposite, the leaves were broadly oval or elliptic, the apex was short or slightly obtuse, the whole or wave-shaped obtuse serrate, the flower yellow, was the head inflorescence. The characters of medicinal materials are yellowish green stems, vertical stripes, section white, hollow, papery leaves, shrinkage, grayish yellow flowers, terminal or axillary. It can be seen that there are 10 ~ 15 outer tough vascular bundles, the pulp is broad and often hollow. In the transverse section of the leaf, the epidermis is not glandular hair and stomata, the main vein vascular bundle is usually 3 bundles; the stomatal axis is infinitive; the pollen grain, non-glandular hair, In the identification of TLC, the TLC of the gold button test sample showed the same blue fluorescence spots in the corresponding position of the control medicine and the reference substance. The water content, total ash content and acid insoluble ash content of gold button medicinal materials shall not be higher than 15.0% and 7.0%, and the content of water soluble extracts shall not be less than 18.0%. The gold button may contain organic acids, flavonoids, phytosterols, coumarins, triterpenes, lactones, etc. Phenols, volatile oil and other chemical constituents, the content of Lv Yuan acid in gold button showed a good linear relationship with the peak area in the range of 0.0100 渭 g, 0.4010 渭 g. The regression equation was yangz2643.72866x-11.165153rn 0.99970.The average recovery rate was 102.82RSD1.900.The Lv Yuan acid content of each batch of gold buttons was between 0.0043 and 0.0857%. The HPLC fingerprint of Jinjiu was preliminarily established, and six common peaks were screened out. Conclusion: the characteristics of gold buttons obtained by pharmacognosy identification are stable and reliable, which can be used as the basis for qualitative identification of gold buttons. Gold button medicine in processing, should pay attention to cleaning, so as not to bring too much sediment, and fully dry, to prevent moldy deterioration, in order to facilitate storage. High performance liquid chromatography (HPLC) was used to determine the content of Lv Yuan acid in gold button. The method is accurate, simple, rapid and reproducible. This study has established the experimental foundation for establishing the quality standard of the Zhuang medicine Jinjiu medicinal material and provided the scientific reference basis for its reasonable development and utilization in the future.
