電針治療小鼠酒精戒斷期抑郁狀態(tài)和機(jī)制研究

【摘要】:目的針灸是祖國(guó)傳統(tǒng)醫(yī)學(xué)的瑰寶,在臨床多種疾病的治療和預(yù)防中有著廣泛的應(yīng)用,電針的發(fā)明為現(xiàn)代醫(yī)學(xué)研究針灸的治療作用和機(jī)制提供了科學(xué)的手段。本實(shí)驗(yàn)研究了2/100Hz電針能否治療小鼠酒精依賴戒斷期的抑郁狀態(tài),能否翻轉(zhuǎn)小鼠酒精依賴戒斷期海馬腦區(qū)腦源性神經(jīng)營(yíng)養(yǎng)因子和神經(jīng)發(fā)生的減少。方法把48只小鼠分為蒸餾水組、酒精依賴戒斷組、束縛組和2/100Hz電針組。首先建立小鼠酒精依賴模型,采用雙瓶自由選擇飲酒的方法進(jìn)行酒精依賴造模,接下來(lái)是14天的自然戒斷,這段時(shí)間內(nèi)給予電針治療。電針組電針每天1次,8:00a.m.,頻率2/100 Hz,強(qiáng)度分別為0.5、1.0、1.5 mA,三個(gè)強(qiáng)度的時(shí)間各為10min;束縛組每天1次,8:00a.m.,30min;自然戒斷組動(dòng)物不做任何處理,常規(guī)飼養(yǎng)。以上各組動(dòng)物每組12只,43天時(shí)全部進(jìn)行強(qiáng)迫游泳實(shí)驗(yàn),檢測(cè)動(dòng)物抑郁狀態(tài),44天時(shí)選擇6只給予BrdU處理(100 mg/kg,i.p.),2小時(shí)之后灌流取材,進(jìn)行免疫組化實(shí)驗(yàn),檢測(cè)神經(jīng)發(fā)生;另外6只直接處死取材,進(jìn)行ELISA實(shí)驗(yàn)檢測(cè)海馬腦源性神經(jīng)營(yíng)養(yǎng)因子含量。結(jié)果1.經(jīng)過(guò)28天的雙瓶自由選擇飲酒實(shí)驗(yàn),檢測(cè)小鼠的酒精偏愛(ài)率和飲酒量,我們的結(jié)果表明每只小鼠的酒精偏愛(ài)率都達(dá)到了75%以上,酒精攝入量都達(dá)到了15g/kg以上,對(duì)酒精形成了明顯的偏愛(ài),而且三組小鼠對(duì)酒精的偏愛(ài)率和酒精攝入量沒(méi)有統(tǒng)計(jì)學(xué)差異(P0.05),同時(shí)我們每周檢測(cè)小鼠體重,結(jié)果表明和蒸餾水組相比,飲酒對(duì)小鼠的體重沒(méi)有影響(P0.05)。2.強(qiáng)迫游泳實(shí)驗(yàn)顯示,酒精戒斷14天后,和蒸餾水組相比,酒精戒斷組的小鼠處于抑郁狀態(tài),強(qiáng)迫游泳的不動(dòng)時(shí)間明顯延長(zhǎng)(P㩳0.01)。ELISA實(shí)驗(yàn)結(jié)果顯示,酒精戒斷后14天,和蒸餾水組相比,戒斷組小鼠海馬腦區(qū)腦源性神經(jīng)營(yíng)養(yǎng)因子的含量明顯減少(P㩳0.05)。BrdU免疫組化結(jié)果顯示,和蒸餾水組相比,酒精戒斷組小鼠海馬神經(jīng)發(fā)生減少(P㩳0.05)。3.在戒斷期給予小鼠2/100Hz電針治療,強(qiáng)迫游泳實(shí)驗(yàn)中可以明顯減少小鼠游泳時(shí)的不動(dòng)時(shí)間(P㩳0.05),表明電針可以改善小鼠酒精依賴戒斷期的抑郁狀態(tài)。ELISA實(shí)驗(yàn)顯示電針可以增加海馬腦區(qū)腦源性神經(jīng)營(yíng)養(yǎng)因子的含量(P㩳0.05),BrdU免疫組化結(jié)果顯示,2/100Hz電針可以促進(jìn)海馬神經(jīng)細(xì)胞發(fā)生的增加(P㩳0.05)。結(jié)論1.雙瓶自由選擇飲酒14天后小鼠表現(xiàn)出抑郁樣行為,減少小鼠海馬腦源性神經(jīng)營(yíng)養(yǎng)因子含量和海馬神經(jīng)發(fā)生。2.2/100Hz電針可以改善酒精依賴戒斷期的抑郁,增加海馬腦源性神經(jīng)營(yíng)養(yǎng)因子含量和神經(jīng)發(fā)生。
[Abstract]:Objective Acupuncture and moxibustion is a treasure of traditional Chinese medicine, which is widely used in the treatment and prevention of many kinds of clinical diseases. The invention of electroacupuncture provides a scientific means for modern medicine to study the therapeutic effect and mechanism of acupuncture and moxibustion. This experiment was conducted to study whether 2/100Hz electroacupuncture could treat depression in alcohol dependence withdrawal period in mice and reverse the decrease of brain-derived neurotrophic factor and neurogenesis in hippocampus of mice during alcohol dependence withdrawal period. Methods 48 mice were divided into distilled water group, alcohol dependent withdrawal group, bound group and 2/100Hz electroacupuncture group. The model of alcohol dependence in mice was established first, and the model was made with the method of double bottles of free choice. Then, the natural withdrawal of 14 days was given, and electroacupuncture was given during this period. In the electroacupuncture group, once a day, 8: 00a.m., with a frequency of 2 / 100 Hz, with the intensity of 0.5 脳 1.0 Ma and 1.5 Ma, the time of the three intensities was 10 min, and the bound group was 8: 00 a.m..30 min. once a day. The animals in the natural abstinence group did not do any treatment and were raised routinely. In each group, 12 animals in each group were subjected to forced swimming for 43 days, and 6 animals were given BrdU for 44 days. After 2 hours of BrdU treatment, the animals were perfused with 100 mg / kg i.p. for 2 hours, and the neurogenesis was detected by immunohistochemistry. The other 6 rats were killed directly and the content of brain-derived neurotrophic factor (BDNF) in hippocampus was detected by ELISA test. Result 1. After 28 days of free choice of alcohol in two bottles, the alcohol preference rate and alcohol consumption of mice were measured. Our results showed that the alcohol preference rate of each mouse was more than 75%, and the alcohol intake was above 15g/kg. There was no significant difference in alcohol preference rate and alcohol intake among the three groups. At the same time, we measured the body weight of mice every week. The results showed that alcohol consumption had no effect on the body weight of mice compared with distilled water group. The forced swimming test showed that after 14 days of alcohol withdrawal, the mice in the alcohol withdrawal group were in a depressed state compared with the distilled water group, and the immobility time of forced swimming was significantly prolonged. The results of Elisa showed that 14 days after alcohol withdrawal, compared with the distilled water group, the immobility time of forced swimming group was significantly longer than that of the distilled water group. The content of brain-derived neurotrophic factor (BDNF) in the hippocampus of abstinent group was significantly decreased. The results of immunohistochemistry showed that compared with distilled water group, the hippocampal neurogenesis of alcohol abstinent group was decreased as compared with that of distilled water group. During the abstinence period, mice were treated with 2/100Hz electroacupuncture, In forced swimming experiment, the immobility time of swimming in mice was significantly reduced, indicating that electroacupuncture could improve the depression state of mice during alcohol dependence withdrawal period. Elisa experiment showed that electroacupuncture could increase brain-derived neurotrophic factor in hippocampal brain area. The results of BrdU immunohistochemistry showed that 2 / 100 Hz electroacupuncture could promote the increase of hippocampal neurogenesis. Conclusion 1. After 14 days of drinking, the mice showed depressive behavior. Reducing the content of brain-derived neurotrophic factor in hippocampus and hippocampal neurogenesis. 2.2 / 100 Hz electroacupuncture could improve depression in alcohol dependence withdrawal period. Increase the content of brain-derived neurotrophic factor and neurogenesis in hippocampus.
