【摘要】：目的探討穴位貼敷治療過敏性哮喘的可能作用機(jī)制。方法將40只Wistar大鼠隨機(jī)分為空白組、模型組、貼敷2 h組、貼敷4 h組,每組10只。除空白組外,其余各組大鼠用雞卵白蛋白(OVA)和生理鹽水的混懸液皮下多點(diǎn)注射及霧化吸入致敏制造過敏性哮喘模型。造模結(jié)束后第15天起,空白組和模型組正常飼養(yǎng);貼敷2 h組、貼敷4 h組于大鼠肺俞、脾俞、腎俞穴位貼敷,分別貼敷2 h、4 h,隔天1次,共貼7次。觀察各組大鼠引喘潛伏期和發(fā)作總持續(xù)時(shí)間;ELISA法檢測(cè)大鼠血清中轉(zhuǎn)化生長(zhǎng)因子β1(TGF-β1)和尾加壓素Ⅱ(UⅡ)含量;HE染色觀察大鼠支氣管和肺組織病理結(jié)構(gòu)改變。結(jié)果與模型組比較,貼敷2 h、4 h組引喘潛伏期延長(zhǎng)、發(fā)作總持續(xù)時(shí)間縮短,血清TGF-β1和UⅡ水平明顯降低(P0.01);支氣管和肺組織結(jié)構(gòu)不清,支氣管黏膜水腫、充血及氣道壁增厚等改變顯著改善。貼敷4 h組與貼敷2 h組相比,引喘潛伏期、發(fā)作總持續(xù)時(shí)間、血清TGF-β1與UⅡ差異均無統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論穴位貼敷能降低血清TGF-β1和UⅡ含量,從而改善氣道重塑,可能是其治療過敏性哮喘的機(jī)制之一。
[Abstract]:Objective to explore the possible mechanism of acupoint application in the treatment of allergic asthma. Methods Forty Wistar rats were randomly divided into blank group, model group, 2 h group and 4 h group with 10 rats in each group. The allergic asthma model was induced by subcutaneous injection of ovalbumin (OVA) and normal saline suspension and atomization inhalation in the other groups except the blank group. From the 15th day after the model was finished, the blank group and the model group were fed normally, while the group of 2 h and the group of 4 h were applied at the acupoints of Fesu, Pi Yu and Shenshu, respectively, for 2 hours and 4 hours respectively, and once every other day for a total of 7 times. The latent period of induced asthma and the total duration of attack were detected by Elisa. The contents of TGF- 尾 1 and U2) in serum of rats were detected. The pathological changes of bronchi and lung were observed by HE staining.
