逆灸對(duì)力竭運(yùn)動(dòng)大鼠心肌組織中AMPK、mTOR的影響

【摘要】:目的:觀測(cè)逆灸對(duì)力竭運(yùn)動(dòng)大鼠心肌組織中一磷酸腺苷活化的蛋白激酶(AMPK)、哺乳動(dòng)物雷帕霉素靶蛋白(mTOR)的影響,探討逆灸抗力竭運(yùn)動(dòng)心肌損傷的作用機(jī)制。方法:將18只雄性SD大鼠隨機(jī)分為空白組、力竭組和逆灸組,每組6只?瞻捉M大鼠每天被操作者用手輕按于操作臺(tái)上,套上特制鼠套,保持5min,不進(jìn)行任何干預(yù);力竭組大鼠前10d不做處理,逆灸組取"足三里""關(guān)元"進(jìn)行干預(yù),每穴每日灸5壯,連續(xù)10d,第11d力竭組和逆灸組大鼠力竭游泳后取材,并記錄兩組大鼠達(dá)到力竭所用時(shí)間。采用HE染色觀察各組大鼠心肌組織炎性反應(yīng);比色法檢測(cè)血清中乳酸脫氫酶(LDH)、超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量;免疫抑制法檢測(cè)血清中肌酸激酶同工酶(CK-MB)活性;Elisa檢測(cè)血清中肌鈣蛋白T(cTnT)含量;Western blot測(cè)定心肌組織中一磷酸腺苷活化的蛋白激酶α2(AMPKα2)、哺乳動(dòng)物雷帕霉素靶蛋白(mTOR)表達(dá)。結(jié)果:逆灸組較力竭組到力竭狀態(tài)所需時(shí)間延長(zhǎng)[(4 831.17±689.88)s vs(3 509.50±1 232.49)s,P0.05],心肌組織結(jié)構(gòu)清楚,炎細(xì)胞浸潤(rùn)減輕;與空白組比較,力竭組血清LDH、CK-MB、cTnT升高(均P0.05);MDA含量升高,SOD活性下降(均P0.05);心肌組織AMPKα2表達(dá)升高、mTOR下降(均P0.05)。與力竭組比較,逆灸組血清LDH、CK-MB、cTnT下降(均P0.05);MDA含量下降,SOD活性升高(均P0.05);心肌組織AMPKα2表達(dá)升高、mTOR下降(均P0.05);AMPKα2與mTOR呈負(fù)相關(guān)(r=-0.764,P0.05)。結(jié)論:逆灸可能通過(guò)調(diào)控AMPK/mTOR表達(dá)誘導(dǎo)相應(yīng)的信號(hào)級(jí)聯(lián)反應(yīng)對(duì)抗一次性力竭運(yùn)動(dòng)大鼠心肌組織損傷。
[Abstract]:Objective: to observe the effect of reverse moxibustion on adenosine monophosphate activated protein kinase AMPKO and mammalian rapamycin target protein mTORin in myocardial tissue of exhaustive exercise rats, and to explore the mechanism of anti-exhaustive exercise myocardial injury by reverse moxibustion. Methods: eighteen male Sprague-Dawley rats were randomly divided into blank group, exhaustive group and reverse moxibustion group with 6 rats in each group. The rats in the blank group were pressed lightly by the operator on the operating platform with their hands every day, the rats in the exhaustive group were not treated for the first 10 days, and the rats in the reverse moxibustion group were treated with "Zusanli" and "Guanyuan" at 5 strong points per day for 5 minutes, and the rats in the exhaustive group were not treated for 5 minutes, and the rats in the reverse moxibustion group were treated with "Zusanli" and "Guan Yuan". The rats in the exhaustion group and the reverse moxibustion group were taken after exhaustive swimming for 10 consecutive days, and the time to reach exhaustion was recorded in the two groups. The myocardial inflammatory reaction was observed by HE staining, the activity of LDH, SOD and the content of malondialdehyde (MDA) in serum were detected by colorimetric method. The activity of creatine kinase isoenzyme (CK-MBB) in serum was determined by immunosuppressive assay. The content of troponin TnT in serum was determined by Western blot. The expression of adenosine monophosphate activated protein kinase 偽 2(AMPK 偽 2 and mammalian rapamycin target protein mTORwere detected by Western blot. Results: the time from exhaustion to exhaustion was prolonged in the reverse moxibustion group [4 831.17 鹵689.88 s vs(3 509.50 鹵1 232.49 s P 0.05], the myocardial tissue structure was clear and the inflammatory cell infiltration was reduced, compared with the blank group. In the exhaustive group, the level of serum AMPK 偽 2 and the expression of AMPK 偽 2 were increased (all P 0.05) and the expression of AMPK 偽 2 was decreased (all P 0.05), while the activity of sod was decreased (P 0.05), and the expression of AMPK 偽 2 was increased (P 0.05). Compared with the exhaustive group, the serum level of AMPK 偽 2 in the moxibustion group was significantly lower than that in the exhaustive group (P 0.05), the activity of AMPK 偽 2 was increased (P 0.05), and the expression of AMPK 偽 2 in myocardium was significantly lower than that in the control group (P 0.05), and there was a negative correlation between AMPK 偽 2 and mTOR (r -0.764P 0.05). Conclusion: moxibustion may induce signal cascade reaction by regulating the expression of AMPK/mTOR against myocardial tissue injury in rats with exhaustive exercise.
