【摘要】：目的:探討滿藥北五味子多糖(NSCP)對脂多糖(LPS)激活的離體培養(yǎng)人中性粒細(xì)胞的影響,闡明其抗炎作用機(jī)制。方法:利用LPS處理人中性粒細(xì)胞建立炎癥細(xì)胞模型,實驗分為對照組、LPS組(1.0mg·L~(-1))及NSCP組(1.25、2.50和5.00g·L~(-1)),NSCP組先加入LPS刺激60min,再加入不同濃度NSCP。應(yīng)用酶聯(lián)免疫吸附法(ELISA)檢測中性粒細(xì)胞中腫瘤壞死因子α(TNF-α)水平,采用流式細(xì)胞術(shù)檢測中性粒細(xì)胞凋亡率。結(jié)果:與對照組比較,LPS組TNF-α水平升高(P0.05);與LPS組比較,NSCP組TNF-α水平降低(P0.05)。LPS組中性粒細(xì)胞凋亡百分率較對照組降低(P0.05);NSCP組中性粒細(xì)胞凋亡百分率隨時間及劑量增加而升高,其中5.00g·L~(-1) NSCP作用16h促進(jìn)其凋亡作用最佳,與LPS組比較凋亡百分率明顯升高(P0.05)。結(jié)論:NSCP可通過抑制LPS誘導(dǎo)TNF-α分泌及中性粒細(xì)胞凋亡延遲而發(fā)揮抗炎作用。
[Abstract]:Aim: to investigate the effect of NSCP on cultured human neutrophils activated by lipopolysaccharide (LPS), and to elucidate its anti-inflammatory mechanism. Methods: LPS was used to treat human neutrophil to establish inflammatory cell model. The model was divided into two groups: control group (LPS group) and NSCP group (1.25g / L ~ 2.50 and 5.00 g / L ~ (-1)) respectively. The cells were stimulated with LPS for 60 min and then added with different concentrations of LPS. The level of tumor necrosis factor- 偽 (TNF- 偽) in neutrophils was detected by Elisa and the apoptosis rate of neutrophils was detected by flow cytometry. Results: compared with the control group, the level of TNF- 偽 increased in LPS group and decreased in LPS group. The percentage of neutrophil apoptosis in NSCP group was higher than that in control group. Among them, 5.00g / L NSCP had the best effect on promoting apoptosis for 16 h, and the percentage of apoptosis in LPS group was significantly higher than that in LPS group (P 0.05). ConclusionTwo-NSCP can inhibit the secretion of TNF- 偽 and delay the apoptosis of neutrophils by inhibiting the secretion of TNF- 偽 by LPS.
