刺糖對(duì)酒精性肝病TNF-α和TLR4表達(dá)及病理變化的影響

【摘要】:探討刺糖對(duì)酒精性肝病(Alcoholic liver disease,ALD)小鼠肝組織TNF-α和TLR4表達(dá)及組織病理變化的影響。將42只小鼠隨機(jī)分成空白組、模型組、水飛薊賓陽(yáng)性組、刺糖高、中、低劑量組。模型組以500mL/L酒精造模,空白對(duì)照組(15mL/kg生理鹽水)、刺糖高、中、低(600、300、150mg/kg)、水飛薊賓(300mg/kg)灌胃,每日藥物處理后用500mL/L酒精(10mL/kg)灌胃2次(分別為藥物處理后2h和藥物處理后10h),連續(xù)3d,末次灌胃酒精12h后,取肝臟提取mRNA,制備肝臟標(biāo)本進(jìn)行HE染色,實(shí)時(shí)熒光定量PCR檢測(cè)肝組織TNF-α和TLR4mRNA表達(dá)。結(jié)果表明,與模型組相比,小鼠肝臟中TNF-α和TLR4mRNA表達(dá)水平明顯降低。各刺糖給藥組均抑制酒精引起的TNF-α和TLR4基因mRNA表達(dá)水平的增強(qiáng)。刺糖高、中、低劑量組均能夠改善酒精所引起的肝臟病理變化?赏瞥龃烫菍(duì)酒精引起的小鼠肝損傷有一定的保護(hù)作用,其作用機(jī)制可能與抑制TNF-α和TLR4基因mRNA表達(dá)有關(guān)。
[Abstract]:To investigate the effect of prickly sugar on the expression of TNF- 偽 and TLR4 in the liver tissue of alcoholic liver disease (ALD) mice and its histopathological changes.42 mice were randomly divided into blank group, model group, silythistle Binyang group, high, middle and low dose groups.The model group was made with 500mL/L alcohol, and the blank control group was treated with 15ml / kg normal saline, 600,300mg / kg silybin and 300mg / kg silybin.Two times (2 h after drug treatment and 10 h after drug treatment) were treated with 500mL/L alcohol (10 mL / kg) for 3 consecutive days. After the last administration of alcohol for 12 h, mRNAs were extracted from the liver, and the liver specimens were prepared for HE staining.The expression of TNF- 偽 and TLR4mRNA in liver tissue was detected by real-time fluorescence quantitative PCR.The results showed that compared with the model group, the expression of TNF- 偽 and TLR4mRNA in the liver of mice decreased significantly.All the prickly sugar groups inhibited the increase of mRNA expression of TNF- 偽 and TLR4 gene induced by alcohol.High, medium and low doses of prickly sugar could improve the pathological changes of liver induced by alcohol.It can be concluded that prickly sugar has a protective effect on the liver injury induced by alcohol in mice, and its mechanism may be related to the inhibition of the expression of TNF- 偽 and TLR4 gene mRNA.
