水藥金耳環(huán)多糖對實驗性2型糖尿病模型大鼠糖脂代謝、腎功能及氧化應(yīng)激的影響

【摘要】:目的:研究水藥金耳環(huán)多糖對實驗性2型糖尿病模型大鼠糖脂代謝、腎功能及氧化應(yīng)激的影響,為金耳環(huán)多糖的開發(fā)利用提供參考。方法:將60只SD大鼠隨機分為正常對照組、模型對照組、陽性對照組(厄貝沙坦片,0.02 g/kg)和金耳環(huán)多糖低、中、高劑量組(以生藥計分別為2、4、8 g/kg),每組10只。除正常對照組外,其余各組大鼠均ip鏈脲佐菌素75 mg/(kg·d)復制糖尿病大鼠模型。成模后,各給藥組大鼠ig相應(yīng)藥物,正常對照組和模型對照組大鼠ig等體積5%羧甲基纖維素鈉溶液,每天2次,連續(xù)42d。給藥結(jié)束后,采用紫外分光光度計檢測肝組織中肝糖原含量;采用全自動生化分析儀檢測大鼠24 h尿量、24 h尿蛋白含量,血清中三酰甘油(TG)、膽固醇(TC)、低密度脂蛋白膽固醇(LDL-C)、高密度脂蛋白膽固醇(HDL-C)、肌酐(Cr)、尿素氮(BUN)水平,以及腎組織中超氧化物歧化酶(SOD)、谷胱甘肽過氧化物酶(GSH-Px)、過氧化氫酶(CAT)、超氧化物歧化酶(ROS)、丙二醛(MDA)水平。結(jié)果:與正常對照組比較,模型對照組大鼠肝組織中肝糖原含量減少(P0.05);24 h尿量、24 h尿蛋白量增加(P0.05);血清中TG、TC、LDL-C水平升高(P0.05),HDL-C水平降低(P0.05);腎組織中SOD、CAT、GSH-Px水平降低(P0.05),ROS、MDA水平升高(P0.05)。與模型組比較,除肝組織中肝糖原含量、血清中HDL-C水平以及金耳環(huán)多糖低劑量組大鼠24 h尿量改善不顯著外,其余各給藥組大鼠上述指標均得到明顯改善(P0.05)。結(jié)論:金耳環(huán)多糖可調(diào)節(jié)實驗性2型糖尿病模型大鼠的血脂代謝紊亂、改善大鼠腎功能和提高腎抗氧化能力。
[Abstract]:Objective: to study the effects of polysaccharides on glucose and lipid metabolism, renal function and oxidative stress in experimental type 2 diabetic rats, and to provide reference for the development and utilization of polysaccharides from gold earrings.Methods: sixty Sprague-Dawley rats were randomly divided into normal control group, model control group, positive control group (Irbesartan 0.02 g 路kg ~ (-1)) and low, medium and high dose group (2: 48 g 路kg ~ (-1) 路kg ~ (-1)) of irbesartan tablets (n = 10).The diabetic rats were induced by streptozotocin (75 mg/(kg d).After the model was established, the rats of each group were given intravenously, and the rats of normal control group and model control group were given 5% sodium carboxymethyl cellulose solution twice a day for 42 days.After administration, the content of liver glycogen in liver tissue was detected by ultraviolet spectrophotometer, and the urine protein content in 24 h urine of rats was detected by automatic biochemical analyzer.Serum triacylglycerol (TGN), cholesterol triglyceride (TC), low density lipoprotein cholesterol (LDL-C), high density lipoprotein cholesterol (HDL-C), creatinine (Cr), urea nitrogen bun (BUNA),And the levels of superoxide dismutase (SOD), glutathione peroxidase (GSH-PxN), catalase (CATX), superoxide dismutase (SOD) and malondialdehyde (MDA) in renal tissue.緇撴灉:涓庢甯稿鐓х粍姣旇緝,妯″瀷瀵圭収緇勫ぇ榧犺倽緇勭粐涓倽緋栧師鍚噺鍑忓皯(P0.05);24 h灝塊噺,24 h灝胯泲鐧介噺澧炲姞(P0.05);琛,
