電針不同穴位組合對功能性腹瀉大鼠空腸腸嗜鉻細胞和5-HT的影響研究

【摘要】:目的:運用一定參數(shù)的電針,干預功能性腹瀉模型大鼠的不同穴位組合,觀察大鼠的各種腹瀉指標及空腸中腸嗜鉻細胞的形態(tài)數(shù)量、5-HT分泌量及TPH的表達量的不同變化,探索電針干預功能性腹瀉的調節(jié)機制,篩選出電針干預功能性腹瀉療效較好的穴位組合。方法:選用20只SD大鼠,隨機分為2組:空白組10只、模型組10只?瞻捉M不做任何處理,模型組給以3ml/只番瀉葉煎液灌胃,1次/天,連續(xù)灌胃14天。灌胃結束后,對兩組大鼠繼續(xù)以正常飼食、飲水,連續(xù)觀察14d,至第28d造模結束,取材,觀察綜合評定各項指標。選用50只SD大鼠,隨機分為5組:空白組、模型組、電針1組、電針2組、電針3組。模型組、電針1組、電針2組、電針3組運用上述腹瀉模型造模方法造模,造模結束后,進行電針干預觀察,空白組、模型組不予以電針,電針組(電針1組、電針2組、電針3組)給予電針干預1個療程(1次/天,10天/療程)。療程結束后,截取大鼠的空腸組織,檢測其腸嗜鉻細胞數(shù)量、5-HT的分泌量及TPH的表達量。結果:番瀉葉煎液灌胃法制備的功能性腹瀉大鼠模型,在通過體重增長率、腹瀉指數(shù)、稀便率、稀便級、浸銀染色等方面的觀察和綜合評定后,證明腹瀉模型大鼠的制備是成功的。造模結束后,與空白組對比,模型組大鼠在空腸部位中的腸嗜鉻細胞數(shù)量和5-HT的分泌量、TPH的表達量均有所升高。電針干預后,與空白組、模型組比較,電針三組均可降低空腸中腸嗜鉻細胞的數(shù)量、5-HT分泌量及TPH的表達量,電針1組(天樞配大腸俞)對空腸腸嗜鉻細胞數(shù)量、5-HT的分泌量、TPH的表達量影響顯著,電針2組(曲池配上巨虛)和電針3組(天樞配大腸俞+曲池配上巨虛)影響不顯著。結論:1.運用番瀉葉煎液灌胃制備的功能性腹瀉大鼠,模型穩(wěn)定,造模成功。2.空腸和回腸是番瀉葉腹瀉模型造模的作用部位,空腸為主要作用部位。3.造模及電針干預過程中,嗜鉻細胞的數(shù)量、5-HT的分泌量及TPH的表達量與功能性腹瀉有相關性聯(lián)系。4.電針干預相關穴位,通過減少腸嗜鉻細胞數(shù)量、5-HT釋放量及TPH表達量,致使信號傳導減弱,起到治療作用。電針1組影響顯著,即天樞配大腸俞為治療功能性腹瀉的較好穴位組合,電針2組和電針3組影響不顯著。
[Abstract]:Objective: to observe the effects of electroacupuncture on different acupoints combination in rats with functional diarrhea, and to observe the changes of various diarrhea indexes and the morphologic quantity of chromaffin cells in jejunum and the expression of TPH. To explore the regulating mechanism of electroacupuncture intervention on functional diarrhea and to screen out the acupoint combination of electroacupuncture for functional diarrhea. Methods: 20 SD rats were selected and randomly divided into 2 groups: blank group (n = 10), control group (n = 10), control group (n = 10), and control group (n = 10). Ten rats in the model group. The blank group was given 3ml/ Senna leaf decoction once a day for 14 days. After the gastric administration, the rats in the two groups continued to be fed with normal diet, drinking water and observed continuously for 14 days until the end of the 28 th day. 50 Sprague-Dawley rats were randomly divided into 5 groups: blank group, model group, electroacupuncture group 1, electroacupuncture group 2, electroacupuncture group 3, model group, electroacupuncture group 1, electroacupuncture group 2. Three groups of electroacupuncture were used to model the diarrhea model. After the model was made, the intervention of electroacupuncture was observed. The blank group and the model group were not given electroacupuncture, and the electroacupuncture group (group 1 and group 2) were treated with electroacupuncture. Electroacupuncture group (group 3) was given electroacupuncture intervention for one course of treatment (10 days per day). After the course of treatment, the jejunum tissue of rats was cut off. Results: the rat model of functional diarrhea was made by gastric perfusion of Senna leaf decoction. Results: the rat model of functional diarrhea was induced by body weight growth rate, diarrhea index, fecal rate and fecal grade. After the observation and comprehensive evaluation of silver staining and other aspects, it was proved that the preparation of diarrhea model rats was successful. After the establishment of the model, it was compared with the blank group. The number of chromaffin cells in jejunum and the secretion of 5-HT in model group were increased. Electroacupuncture could decrease the secretion of 5-HT and the expression of TPH in jejunum. The effect of electroacupuncture on the secretion of 5-HT and the expression of 5-HT in jejunal chromaffin cells was significant in group 1 (Tianshu combined with Daichang Shu). The effects of electroacupuncture group 2 (zhuchi plus great asthenia) and electroacupuncture group 3 (Tianshu group combined with Dachang Yu koji group on great asthenia) were not significant. Conclusion: 1. The model of functional diarrhea rats prepared by gastric perfusion with Senna leaf decoction was stable. 2. Jejunum and ileum are the acting sites of the model of Senna leaf diarrhea, and jejunum is the main part of the model. 3. In the process of model making and electroacupuncture intervention, The number of chromaffin cells and the expression of TPH were related to the functional diarrhea. 4. Electroacupuncture interfered with the related acupoints. By reducing the number of chromaffin cells and the expression of TPH, the signal transduction was weakened. The effect of electroacupuncture group 1 was significant, that is, Tianshu combined with Dachang Shu was a better combination of acupoints for the treatment of functional diarrhea, but the effect of electroacupuncture group 2 and electroacupuncture group 3 was not significant.
