壯醫(yī)復(fù)方蓮素飲對圍絕經(jīng)期模型大鼠骨代謝相關(guān)因子影響的實驗研究

【摘要】:目的:觀察壯醫(yī)復(fù)方蓮素飲對圍絕經(jīng)期模型大鼠骨代謝相關(guān)因子的影響,探討壯醫(yī)復(fù)方蓮素飲對圍絕經(jīng)期綜合征模型大鼠骨代謝的作用機(jī)理。方法:選擇成年雌性SD大鼠,腹腔注射4-VCD芝麻油溶劑造模15天,4-VCD劑量為80 mg/kg。選擇造模成功大鼠50只,隨機(jī)分為5組,每組10只,分別為復(fù)方蓮素飲高、中、低劑量組、模型組、補(bǔ)佳樂組,另選未造模的10只作為正常對照組。對照組、模型組給予蒸餾水灌胃,復(fù)方蓮素飲高、中、低劑量組分別按5.76g/kg、2.88g/kg、1.44g/kg劑量給予灌胃,補(bǔ)佳樂組按0.09mg/kg給予灌胃,各組連續(xù)灌胃給藥21天。給藥結(jié)束后,取樣本,測定血清中E2、LH、FSH,降鈣素基因相關(guān)肽(CGRP)、血管活性腸肽(VIP)的含量;子宮和卵巢組織中基質(zhì)Gla蛋白(MGP)的表達(dá)。結(jié)果:壯醫(yī)復(fù)方蓮素飲能顯著提升圍絕經(jīng)期模型大鼠血清中E2水平,與模型組相比,復(fù)方蓮素飲低、中劑量組的E2水平升高,差異具有統(tǒng)計學(xué)意義(P0.05),補(bǔ)佳樂組與復(fù)方蓮素飲高劑量組的E2水平明顯升高,差異均具有顯著統(tǒng)計學(xué)意義(P0.01);并降低血清中FSH、LH水平,與模型組相比,復(fù)方蓮素飲低、中劑量組的FSH、LH水平下降,差異具有統(tǒng)計學(xué)意義(P0.05),補(bǔ)佳樂組與復(fù)方蓮素飲高劑量組的FSH、LH水平明顯下降,差異均具有顯著統(tǒng)計學(xué)意義(P0.01);降低血清中CGRP、VIP水平,與模型組相比,復(fù)方蓮素飲中劑量組的CGRP、VIP水平下降,差異具有統(tǒng)計學(xué)意義(P0.05),補(bǔ)佳樂組與復(fù)方蓮素飲高劑量組的CGRP、VIP水平明顯下降,差異均具有顯著統(tǒng)計學(xué)意義(P0.01);顯著提升子宮、卵巢組織中MGP的表達(dá),與模型組相比,復(fù)方蓮素飲中劑量組的MGP基因表達(dá)水平升高,差異具有統(tǒng)計學(xué)意義(P0.05),補(bǔ)佳樂組與復(fù)方蓮素飲高劑量組的MGP基因表達(dá)水平明顯升高,差異均具有顯著統(tǒng)計學(xué)意義(P0.01)。結(jié)論:壯醫(yī)復(fù)方蓮素飲通過提升E2水平,降低LH、FSH水平,調(diào)控生殖軸,平衡機(jī)體紊亂的性激素;下調(diào)CGRP、VIP表達(dá)水平,改善骨代謝和血管舒縮功能,緩解因圍絕經(jīng)期雌激素水平下降引起的骨流失加速和骨合成能力下降等一系列骨代謝相關(guān)情況,以及血管舒張過度引起的機(jī)體潮熱汗出癥狀;上調(diào)MGP表達(dá)水平,促進(jìn)骨生成抑制骨破壞,改善異常骨代謝。綜上,本方能夠改善圍絕經(jīng)期綜合征因激素神經(jīng)內(nèi)分泌紊亂而出現(xiàn)的潮熱、骨代謝異常等情況,起到調(diào)理內(nèi)分泌、調(diào)節(jié)血管舒縮,改善骨代謝的作用。
[Abstract]:Objective: to observe the effect of Lianxin decoction on bone metabolism related factors in peri-menopausal rats, and to explore the mechanism of bone metabolism in rats with perimenopausal syndrome. Methods: adult female SD rats were selected. The dose of 4-VCD was 80 mg / kg / kg for 15 days by intraperitoneal injection of 4-VCD sesame oil solvent. Fifty successful rats were randomly divided into 5 groups, 10 rats in each group, respectively: compound Lianyin high, middle and low dosage groups, model group and Bujiale group. Another 10 rats were selected as normal control group. The model group was given distilled water by gastric perfusion. The group of high, middle and low dosage of compound Liansu drink was given intragastric perfusion at the dose of 5.76 g / kg ~ 2.88 g / kg ~ (1.44) g / kg, and Bujiale group was given a dose of 0.09 mg / kg 路kg ~ (-1) 路kg ~ (-1) 路kg ~ (-1) 路kg ~ (-1) 路kg ~ (-1), respectively. Each group was given orally for 21 days. After the administration, samples were taken to determine the contents of serum E _ 2 LHH _ (FSH), calcitonin gene-related peptide (CGRPN) and vasoactive intestinal peptide (VIP). Results: the serum E _ 2 level of the model rats was significantly increased by Liansu decoction, compared with the model group, and the level of E _ 2 in the middle dose group was higher than that in the model group. The difference was statistically significant (P 0.05), the level of E _ 2 in the Bujiale group and the high dose group of compound Liansu decoction were significantly increased, the difference was statistically significant (P < 0.01), and the level of FSHN LH in serum was decreased, compared with the model group, the level of compound Lianyin drink was lower than that in the model group. The level of FSHP LH in the middle dose group decreased significantly (P 0.05), and the level of FSHP LH in the Bujiale group and the high dose group of Fufang Liansu Yin decreased significantly (P 0.01), and the level of CGRP VIP in the serum decreased, compared with the model group, the level of FSHP LH in the middle dose group was significantly lower than that in the model group. The level of CGRPU VIP in the middle dose group of Fufang Liansu decoction decreased, the difference was statistically significant (P 0.05), and the level of VIP in the Bujiale group and the high dose group of Fufang Liansuyin decoction decreased significantly, and the difference was statistically significant (P 0.01). Compared with the model group, the expression level of MGP gene in the middle dose group of Fufang Liansu decoction was higher than that in the model group, and the difference was statistically significant (P 0.05). The expression level of MGP gene in the Bujiale group and the high dose group of Fufang Liansuyin group was significantly higher than that in the model group. Conclusion: Liansu decoction can improve the level of E2, decrease the level of LHH FSH, regulate the reproductive axis, balance the sex hormone of body disorder, down-regulate the expression of CGRPnP VIP, improve bone metabolism and vasomotor function. To relieve a series of bone metabolism related conditions, such as bone loss acceleration and bone synthesis ability decline caused by the decline of estrogen level in perimenopausal period, as well as the symptoms of hot sweating caused by excessive vasodilation, and to upregulate the expression of MGP. To promote bone formation, inhibit bone destruction and improve abnormal bone metabolism. In summary, the prescription can improve the hot flashes and abnormal bone metabolism in peri-menopausal syndrome due to hormonal neuroendocrine disorders, and regulate endocrine, vasomotor and contraction, etc. The effect of improving bone metabolism.
