基于數(shù)據(jù)挖掘和網(wǎng)絡(luò)藥理學(xué)的兒童食積胃腸積熱證組方用藥規(guī)律及作用機(jī)制分析

【摘要】:目的:挖掘兒童食積胃腸積熱證的組方用藥規(guī)律,分析處方可能存在的藥物靶標(biāo)及相互作用,探索核心藥物的作用機(jī)制。方法:收集CNKI中關(guān)于兒童食積胃腸積熱證的病例報道文獻(xiàn),采用TCMISS軟件分析藥物間的關(guān)聯(lián)規(guī)則,挖掘核心藥物及新處方;運用SPSS軟件對藥物的性味歸經(jīng)進(jìn)行系統(tǒng)聚類分析;運用TCMSP、String及KEGG數(shù)據(jù)庫分析新處方各藥物靶標(biāo)間相互作用關(guān)系及相關(guān)通路。結(jié)果:在篩選出的150首處方中,挖掘出6個核心藥物組合和3首新處方,總結(jié)出性味歸經(jīng)相近的9個高頻藥群,得到新處方中相關(guān)藥物靶標(biāo)330個,這些靶標(biāo)構(gòu)成的網(wǎng)絡(luò)中共存在相互作用2 961項、相關(guān)通路137條(FDR0.01),其中與消化系統(tǒng)關(guān)系最密切的7條。結(jié)論:治療兒童食積胃腸積熱證以消積導(dǎo)滯、理氣健脾、清熱除濕為主,其機(jī)制可能通過調(diào)控消化液的分泌,調(diào)控腸道消化及免疫、機(jī)體對微生物的免疫功能等多靶標(biāo)復(fù)雜通路的共同干預(yù)實現(xiàn)。
[Abstract]:Objective: to explore the prescriptions of children with gastroenteric heat accumulation syndrome and analyze the possible drug targets and their interactions in the prescriptions. Methods: to investigate the mechanism of action of core drugs. Methods: collecting the case reports of children's gastroenteric heat syndrome in CNKI, using TCMISS software to analyze the association rules between drugs, excavating the core drugs and new prescriptions; SPSS software was used for systematic cluster analysis of drug quality and flavor, and TCMSPN String and KEGG database were used to analyze the interaction between drug targets and related pathways. Results: among the 150 selected prescriptions, Six core drug combinations and three new prescriptions were excavated, 9 high frequency drug groups with similar properties were summarized, 330 drug targets were obtained, and 2,961 interactions were found in the network composed of these targets. There are 137 related pathways of FDR 0.01, of which 7 are most closely related to digestive system. Conclusion: the treatment of children's syndrome of stomach and intestines accumulation and heat accumulation is mainly composed of eliminating accumulation and stagnation, regulating qi and invigorating spleen, clearing heat and dehumidification, and its mechanism may be through regulating the secretion of digestive fluid. Regulation of intestinal digestion and immunity, immune function of microorganism and other multi-target complex pathways to achieve joint intervention.
