針刺對(duì)血管性癡呆大鼠海馬Ref-1上下游調(diào)控分子的影響

【摘要】:目的:觀察針刺對(duì)血管性癡呆大鼠海馬Ref-1上下游調(diào)控分子的影響,探討針刺基于Ref-1信號(hào)通路的抗氧化機(jī)制。方法:Wistar大鼠82只,隨機(jī)分為假手術(shù)組、模型組、針刺組和非穴組。雙側(cè)頸總動(dòng)脈阻斷法復(fù)制血管性癡呆大鼠模型,造模后第3天針刺,針刺治療結(jié)束后,采用Morris水迷宮檢測(cè)大鼠認(rèn)知功能;尼氏染色檢測(cè)大鼠海馬神經(jīng)元凋亡情況;Western blot技術(shù)檢測(cè)大鼠海馬磷酸化CKⅡ蛋白表達(dá)水平;免疫組化技術(shù)檢測(cè)大鼠海馬HIF-1α蛋白表達(dá)情況。結(jié)果:與假手術(shù)組比較,造模后大鼠認(rèn)知功能受損,海馬區(qū)神經(jīng)元凋亡,磷酸化CKⅡ水平顯著下降(P0.05),HIF-1α表達(dá)水平顯著升高(P0.05)。針刺組大鼠學(xué)習(xí)記憶能力及神經(jīng)元凋亡情況顯著改善,磷酸化CKⅡ水平顯著升高(P0.05),HIF-1α表達(dá)水平顯著降低(P0.05),且改善程度顯著優(yōu)于非穴組(P0.05)。結(jié)論:針刺通過(guò)提高海馬Ref-1上游CKⅡ的磷酸化表達(dá)水平,降低海馬Ref-1下游HIF-1α表達(dá)水平,發(fā)揮抗氧化作用,抑制神經(jīng)元細(xì)胞凋亡,改善學(xué)習(xí)記憶能力。
[Abstract]:Objective: to observe the effect of acupuncture on the upstream and downstream regulation molecules of Ref-1 in hippocampus of vascular dementia rats, and to explore the antioxidant mechanism of acupuncture based on Ref-1 signaling pathway. Methods: 82 Wistar rats were randomly divided into sham operation group and model group. The rat model of vascular dementia was established by bilateral common carotid artery occlusion. After acupuncture on the 3rd day after acupuncture, the cognitive function of the rats was measured by Morris water maze. The apoptosis of hippocampal neurons in rats was detected by Nissl staining.
