【摘要】：目的:通過觀察針刺聯(lián)合亞低溫對腦缺血再灌注損傷(CIRI)模型大鼠神經(jīng)功能缺損評分、梗死面積比、Mi RNA的表達(dá)的影響,探討針刺聯(lián)合亞低溫是否能通過調(diào)控CIRI大鼠的相關(guān)Mi RNA功能,產(chǎn)生腦保護(hù)效應(yīng)。方法:將60只SD大鼠隨機(jī)分為空白組、假手術(shù)組、模型組、針刺組、亞低溫組、針刺聯(lián)合亞低溫組,參照Zea Longa線拴法并加以改良,復(fù)制大腦中動脈缺血模型(MCAO),針刺組、亞低溫組、聯(lián)合組做相應(yīng)治療,治療72h后,用基因微陣列技術(shù)篩選差異表達(dá)mi RNA,對比其功能,并于對大鼠行神經(jīng)功能缺損評分、TTC染色檢測的梗死面積比進(jìn)行比較。結(jié)果:1、各組大鼠神經(jīng)功能缺損評分的比較:治療前,與空白組及假手術(shù)組比較,針刺組、亞低溫組、針刺聯(lián)合亞低溫組神經(jīng)功能缺損評分均降低,差異具有統(tǒng)計學(xué)意義(P0.05或P0.01);治療后,與空白組及假手術(shù)組比較,模型組、針刺組、亞低溫組神經(jīng)功能缺損評分較高,差異具有顯著統(tǒng)計學(xué)意義(P0.01),與模型組比較,針刺組、亞低溫組、針刺聯(lián)合亞低溫組均能降低神經(jīng)功能缺損評分,差異具統(tǒng)計學(xué)意義(P0.05或P0.01),各治療組之間相比,差異無統(tǒng)計學(xué)意義(P0.05)。2、各組大鼠梗死面積比的比較:與空白組及假手術(shù)組比較,模型組、針刺組、亞低溫組及針刺聯(lián)合亞低溫組梗死面積比均明顯增加,差異有統(tǒng)計學(xué)意義(P0.01);與模型組比較,針刺組、亞低溫組及針刺聯(lián)合亞低溫組梗死面積比均減少,差異有統(tǒng)計學(xué)意義(P0.05或P0.01)。3、與空白組比較,模型組差異表達(dá)mi RNAs個數(shù)為4;其功用參與細(xì)胞增殖和細(xì)胞凋亡調(diào)控;與模型組相比:差異表達(dá)mi RNAs數(shù),針刺組為16、亞低溫組為7,針刺聯(lián)合亞低溫組為23;其功用都參與調(diào)節(jié)細(xì)胞增殖、基因表達(dá)、細(xì)胞凋亡、神經(jīng)元分化、軸突延伸、炎性因子產(chǎn)生,且參與每項功用的mi RNAs數(shù)目均不同。結(jié)論:腦缺血再灌后有多種mi RNAs功用失調(diào),針刺聯(lián)合亞低溫可以改善神經(jīng)功能,保護(hù)腦組織,其作用機(jī)制可能與多方面、多環(huán)節(jié)調(diào)節(jié)相關(guān)mi RNAs的功用有關(guān)。
[Abstract]:Objective: to observe the effect of acupuncture combined with mild hypothermia on neurological impairment score and infarct area ratio of Mi RNA in rats with cerebral ischemia-reperfusion injury. To investigate whether acupuncture combined with mild hypothermia can produce brain protective effect by regulating the function of Mi RNA in CIRI rats. Methods: sixty SD rats were randomly divided into blank group, sham operation group, model group, acupuncture group and mild hypothermia group. Acupuncture combined with mild hypothermia group, referring to the method of Zea Longa thread tying and improving, the model of middle cerebral artery ischemia (MCAO), acupuncture group, mild hypothermia group, combined group were treated with corresponding therapy after 72 hours of treatment. The differentially expressed mi RNAs were screened by gene microarray technique, and their functions were compared, and the infarct area ratio detected by TTC staining was compared in rats. Results: 1, comparison of neurological deficit scores in each group: before treatment, Compared with the blank group and sham operation group, the scores of nerve function defect in acupuncture group, mild hypothermia group, acupuncture combined with hypothermia group were all decreased, the difference was statistically significant (P0.05 or P0.01), after treatment, compared with blank group and sham operation group, model group, acupuncture group, Compared with the model group, acupuncture group, mild hypothermia group and acupuncture combined with hypothermia group all decreased the neurological function defect score. The difference was statistically significant (P 0.05 or P 0.01). There was no significant difference between the treatment group and the treatment group. The ratio of infarct area in each group was: compared with the blank group and sham operation group, the model group, the acupuncture group, the control group, the control group, the acupuncture group, the control group, the model group, the acupuncture group, the control group, the model group and the acupuncture group. The infarct area ratio of mild hypothermia group and acupuncture combined with mild hypothermia group was significantly increased, the difference was statistically significant (P 0.01), compared with model group, the infarct area ratio of acupuncture group, mild hypothermia group and acupuncture combined with mild hypothermia group were all decreased. Compared with the blank group, the number of RNAs expressed in the model group was 4; its function was involved in the regulation of cell proliferation and apoptosis; compared with the model group, the number of the difference expressed mi RNAs was higher than that in the model group. 16 in acupuncture group, 7 in mild hypothermia group and 23 in acupuncture combined with mild hypothermia group. Their functions were involved in regulating cell proliferation, gene expression, apoptosis, neuronal differentiation, axon extension and inflammatory factor production. The number of mi RNAs involved in each function is different. Conclusion: there are many kinds of RNAs dysfunction after cerebral ischemia reperfusion. Acupuncture combined with mild hypothermia can improve nerve function and protect brain tissue. Multi-link regulation related to the function of the relevant mi RNAs.
