電針調(diào)控新西蘭兔骨關(guān)節(jié)炎性疼痛及對(duì)脊髓IL-17、IL-17R表達(dá)的影響

【摘要】:目的研究電針是否通過抑制脊髓內(nèi)白細(xì)胞介素(IL)-17及IL-17R的表達(dá)來調(diào)控新西蘭兔骨關(guān)節(jié)炎性疼痛。方法新西蘭兔隨機(jī)分為假手術(shù)組、炎癥模型組、假手術(shù)+IL-17組、炎癥+抗IL-17組、炎癥+電針組、炎癥+假電針組,每組8只,4%木瓜蛋白酶構(gòu)建兔骨關(guān)節(jié)炎模型,IL-17及IL-17抗血清采用椎管內(nèi)注射,電針組選擇電針刺激足三里(30min,2 Hz連續(xù)波,1~2mA),假電針組采用非穴位刺激,測(cè)定兔痛閾,采用實(shí)時(shí)熒光定量PCR(RT-qPCR)檢測(cè)兔脊髓組織IL-17、IL-17R的mRNA表達(dá),Western bolt檢測(cè)IL-17、IL-17R蛋白表達(dá)。結(jié)果在構(gòu)建骨關(guān)節(jié)炎模型后新西蘭兔痛閾顯著下降(P0.05),椎管注射IL-17后可以顯著降低假手術(shù)新西蘭兔痛閾(P0.05),而椎管注射IL-17抗血清后可明顯增加骨關(guān)節(jié)炎模型兔痛閾值。選擇足三里電針刺激可顯著提升兔痛閾(P0.05)。骨關(guān)節(jié)炎模型組IL-17、IL-17R的mRNA及蛋白表達(dá)顯著增加(P0.05),而電針刺激后其表達(dá)顯著下降(P0.05)。結(jié)論新西蘭兔骨關(guān)節(jié)炎性疼痛發(fā)生可能涉及脊髓IL-17異常表達(dá),而電針可能通過抑制脊髓IL-17、IL-17R的表達(dá)來調(diào)控骨關(guān)節(jié)炎性疼痛。
[Abstract]:Objective to study whether electroacupuncture regulates osteoarthritis pain in New Zealand rabbits by inhibiting the expression of interleukin-17 and IL-17R in spinal cord. Methods New Zealand rabbits were randomly divided into three groups: sham operation group, inflammatory model group, IL-17 group and anti-inflammatory IL-17 group. Inflammatory electroacupuncture group, inflammatory pseudoelectroacupuncture group, 8 rabbits in each group with 4% papain to construct rabbit osteoarthritis model, IL-17 and IL-17 antiserum were injected into the spinal canal. Electroacupuncture group (EA group) was used to stimulate Zusanli (Zusanli) for 30 min (2 Hz) and sham electroacupuncture group (sham group) was used to measure the pain threshold of rabbits by non-acupoint stimulation. The expression of IL-17hIL-17R in rabbit spinal cord tissue was detected by real-time fluorescence quantitative PCR- RT-qPCR.The expression of IL-17hIL-17R protein in rabbit spinal cord tissue was detected by Western bolt. Results the pain threshold of New Zealand rabbit decreased significantly after the establishment of osteoarthritis model (P 0.05), and the spinal canal injection of IL-17 could significantly reduce the new sham-operation. The pain threshold of New Zealand rabbit was P0.05A, and the pain threshold of rabbit model of osteoarthritis was significantly increased after injecting IL-17 antiserum into the spinal canal. Electroacupuncture stimulation of Zusanli could significantly increase the pain threshold of rabbit. The expression of mRNA and protein of IL-17 and IL-17R in osteoarthritis model group increased significantly (P 0.05). Conclusion the occurrence of osteoarticular inflammatory pain in New Zealand rabbits may involve abnormal expression of IL-17 in spinal cord. Electroacupuncture may regulate osteoarthritis pain by inhibiting the expression of IL-17 and IL-17 R in spinal cord.
