損傷血瘀證大鼠血清TNF-α、β及骨骼肌細胞凋亡的變化研究

【摘要】:目的:課題從損傷大鼠血清TNF-α與TNF-β以及骨骼肌細胞凋亡變化入手,觀察TNF-α與TNF-β以及骨骼肌細胞凋亡在不同時相的變化規(guī)律,從而分析TNF-α、TNF-β、骨骼肌細胞凋亡變化的特征,部分揭示損傷血瘀證骨骼肌細胞的病理過程。方法:80只Wistar大鼠SPF級別,體重200-250g,每組雌雄各半隨機分為8組。隨機設(shè)立對照組C組(n=10只,下同),傷后1小時組(1H組),傷后12小時組(12H組),傷后1天組(1D組),傷后3天組(3D組),傷后5天組(5D組),傷后7天組(7D組),傷后14天組(14D組),采用“定量化重力擊打法”[1]造成大鼠左大腿內(nèi)群肌肉損傷,并選取大鼠血清以及左大腿內(nèi)群肌肉,用ELISA法測定各組TNF-α與TNF-β的變化。用TUNEL法測定各組的骨骼肌細胞凋亡情況。應用SPSS17.0統(tǒng)計軟件進行相關(guān)的統(tǒng)計方法分析。結(jié)果:1.在骨骼肌細胞凋亡變化方面,1D組、3D組、5D組、7D組、14D組比C組有極顯著升高(P0.01)。3D組、5D組、7D組、14D組比1H組有極顯著升高(P0.01),1D組比1H組有顯著升高(P0.05)。3D組、5D組、7D組、14D組比12H組有極顯著升高(P0.01),1D組比12H組有顯著升高(P0.05)。1D組比C組有極顯著升高(P0.01),比1H組、12H組有顯著升高(P0.05)。7D組比C組、1H組、12H組有極顯著升高(P0.01),比1D組有顯著升高(P0.05)。2.在TNF-α數(shù)量變化方面,12H組比C組有極顯著升高(P0.01),1H組比C組有顯著升高(P0.05)。1H組比C組、14D組有顯著升高(P0.05)。12H組比C組、14D組有極顯著升高(P0.01),12H組比1D組、5D組、7D組TNF-α數(shù)量有顯著升高(P0.05)。3.在TNF-β數(shù)量變化方面,12H組、1D組比C組有極顯著升高(P0.01),1H組比C組有顯著升高(P0.05)。4.TNF-α與TNF-β在本次研究中相關(guān)性低(P≥0.05);TNF-α與TNF-β的比值與反比值無明顯統(tǒng)計學差異(P0.05)。結(jié)論:1.損傷血瘀證病理過程中骨骼肌細胞凋亡的變化在損傷后24小時最為顯著。2.損傷血瘀證中大鼠血清TNF-α的波動變化主要發(fā)生在損傷后24小時之內(nèi),損傷后12小時TNF-α的測試值達到高峰;可能提示TNF-α是損傷血瘀證骨骼肌細胞凋亡發(fā)生的誘導因素之一。3.損傷血瘀證中大鼠血清TNF-β的上升波動變化主要發(fā)生在24小時之內(nèi);這可能也是激發(fā)損傷血瘀證骨骼肌細胞凋亡的重要因素。4.TNF-α與TNF-β之間未呈現(xiàn)線性相關(guān)關(guān)系,其比值與反比值在損傷過程中的不同時間點未產(chǎn)生顯著的變化,即保持相對穩(wěn)態(tài),說明不同時間點TNF-α與TNF-β的偏移并沒有打破二者之間的比值平衡關(guān)系,這有可能類似于中醫(yī)所說的陰陽平衡關(guān)系,因此陰性結(jié)論也有進一步科學探索的價值。
[Abstract]:Objective: to observe the changes of TNF- 偽 and TNF- 尾 and skeletal muscle cell apoptosis in different phases from the changes of serum TNF- 偽 and TNF- 尾 and skeletal muscle cell apoptosis in injured rats, so as to analyze the characteristics of TNF- 偽 TNF- 尾 and skeletal muscle cell apoptosis. Methods 80 Wistar rats, weighing 200-250 g, were randomly divided into 8 groups. 1 hour after injury, 1H group, 12 hours after injury, 12 hours after injury, 1 day after injury, 1 day after injury, 3 days after injury, 5 days after injury, 5 days after injury, 5 days after injury, 7 days after injury, 7 days after injury, 7 days after injury, 14 days after injury, 14 days after injury, 14 days after injury, 14 days after injury, 14 days after injury. The method [1] caused muscle damage in the left thigh of rats. Serum and left thigh muscles were selected. The changes of TNF- 偽 and TNF- 尾 in each group were measured by ELISA method. Apoptosis of skeletal muscle cells in each group was determined by TUNEL method. The correlation statistical analysis was carried out by SPSS17.0 statistical software. Results 1. The P0.01D group was significantly higher than that of the 1H group, the P0.05D group was significantly higher than the 1H group, the P0.05D group was significantly higher than that of the 1H group, and the P0.01D group was significantly higher than that of the 12H group. There was a very significant increase in the P0.01D group compared with the 12H group in the 7D group and the 14D group in the 7D group compared with the 12H group. There was a very significant increase in the P0.01D group compared with the 12H group in P0.05D group. 1D group in the 1D group was significantly higher than that in the 12H group, and the value of P0.05D group was significantly higher than that in the 12H group. Compared with group C, group C had a very significant increase in P0.01D, and group 12D in group 1H was significantly higher than group C in P0.05, group 7D, group C, group C, group 1H, group 12H, and group C, group C, the level of P0.05A was significantly higher than that in group C, P0.05n.2.The quantity of TNF- 偽 in 12H group was significantly higher than that in group C, and that in group 12H was significantly higher than that in group C. There was a significant increase in the number of TNF- 偽 in group P0.05U 路1H than in group C (P 0.05U. 12H) than in group C (group C) 14D. The amount of TNF- 偽 in group 7D was significantly higher than that in group C (P 0.01D), and the change of TNF- 尾 in group 12H was significantly higher than that in group C (P 0.01 ~ 1H), and the level of TNF- 偽 in group 12H was significantly higher than that in group C (P < 0.05), and the level of TNF- 偽 in group C was significantly higher than that in group C (P < 0.05), and the level of TNF- 偽 in group C was significantly higher than that in group C.
