丹參多酚酸鹽干預非ST段抬高心肌梗死患者microRNA與靶基因

【摘要】:目的:高通量檢測非ST段抬高性心肌梗死(NSTEMI)患者藥物干預前后的微小RNA(microRNA)表達譜,對比中西藥物治療前后microRNA表達變化,分析活血化瘀藥物干預靶標與機制。方法:選取符合西醫(yī)疾病診斷標準和中醫(yī)證侯診斷標準的患者,分為2組,包括NSTEMI中藥治療組,NSTEMI西藥組,每組納入35例,抽取外周血,分離外周血單核細胞(PBMC),提取總RNA。根據(jù)各組差異表達的microRNA和基因的通路和網(wǎng)絡(luò)分析結(jié)果,確定各組中起關(guān)鍵調(diào)控作用的microRNA及其靶基因。結(jié)果:在microRNA表達譜中,中藥治療組治療前后、中藥治療組與西藥組兩組治療后比較均存在差異;中藥治療組治療后hsa-miR-92a,hsa-miR-363,hsa-miR-499,hsa-miR-30b,hsa-miR-454-3p,hsa-miR-933表達下調(diào),hsa-miR-144,hsa-miR-451,hsa-miR-494,hsa-miR-320表達上調(diào)。表明這10個microRNA是丹參多酚酸鹽干預NSTEMI的靶標。實時熒光定量PCR(Real-time PCR)驗證,與西藥組比較,中藥治療組治療后hsa-miR-92a,hsa-miR-30b,hsa-miR-499,hsa-miR-454表達下調(diào);中藥治療組治療后hsa-miR-494的表達上調(diào)(P0.05)。結(jié)論:NSTEMI患者治療前后,中藥治療組與西藥組microRNA的表達譜均存在差異,篩選的miRNA可能是活血化瘀藥物干預的靶標。
[Abstract]:Objective: to detect the microRNAs expression profile of non-ST-segment elevation myocardial infarction (NSTEMI) patients before and after drug intervention. Compare the changes of microRNA expression before and after western medicine treatment, analyze the intervention target and mechanism of drugs for promoting blood circulation and removing blood stasis. Methods: select the patients who accord with the diagnostic criteria of western medicine and TCM syndromes. The patients were divided into two groups, including NSTEMI treated group with NSTEMI western medicine group, each group included 35 cases. Peripheral blood was extracted and peripheral blood monocytes were isolated. Total RNA was extracted. The results of microRNA and gene network analysis were analyzed according to the differentially expressed microRNA and gene in each group. Results: in the microRNA expression profile, the traditional Chinese medicine treatment group before and after treatment. There were differences between the Chinese medicine treatment group and the western medicine group after treatment. In the treatment group, hsa-miR-92a, hsa-miR-363, hsa-miR-499and hsa-miR-30b were used. The expression of hsa-miR-454-3psa-miR-933 was down-regulated by hsa-miR-144, hsa-miR-451and hsa-miR-494. The expression of hsa-miR-320 was up-regulated, which indicated that these 10 microRNA were the targets of Salvia miltiorrhiza polyphenolic acid to interfere with NSTEMI. Real-time PCR) validation. Compared with the western medicine group, the expression of hsa-miR-92ahsa-miR-30bsa-miR-499nhsa-miR-454 was down-regulated in the traditional Chinese medicine treatment group. The expression of hsa-miR-494 was up-regulated in the Chinese medicine treatment group after treatment. Conclusion: before and after treatment, the expression of hsa-miR-494 in the treatment group is higher than that in the control group. There were differences in microRNA expression profiles between Chinese medicine group and western medicine group. The selected miRNA might be the target of blood circulation and blood stasis drug intervention.
