【摘要】：研究目的通過動物實驗,本研究觀察針刺“足三針”穴組對大鼠腹部術后結腸Cajal細胞修復微環(huán)境中CSE表達的影響,深入探究針刺對胃腸動力障礙的影響和機制研究,并進一步深入了解針灸在腹部術后胃腸動力障礙功能的恢復方面提供了一定的理論依據(jù)。研究方法在相同實驗條件下,將30只SPF級成年健康SD大鼠隨機分為10只正常組、10只模型組、10只針刺組。取模型組和針刺組大鼠,術前禁食12h,10%水合氯醛行腹腔注射麻醉(按照330ml/kg),實驗大鼠實施結腸吻合術,做成胃腸動力障礙結腸術后模型。針刺組予術后次日起每日給予針刺足三針穴組,模型組和針刺組保持同時進行,每天同一時間行空抓15min,來降低人為固定的影響因素。正常組不予處理。連續(xù)干預3天。期間,記錄好每天各老鼠的進食量、大便顆粒和風干后大便的質量。實驗結束后,觀察小腸推進率并取各組大鼠結腸(盲腸后約2cm處),將取出的組織稱量、標記、記錄質量,盡快放入-80℃冰箱中保存,隨后進行CSE表達的檢測。研究結果1.測定術后大鼠胃腸道排空30min后小腸推進率的情況,與正常組相比,模型組術后小腸推進率較正常組顯著下降(P＜0.05),針刺組比模型組小腸推進率顯著增加,其差異具有統(tǒng)計學意義(P＜0.05)。實驗表明針刺對腹部術后大鼠胃腸動力障礙具有改善作用。2.結腸CSE活性在術后有了大幅度的提升,模型組大鼠結腸CSE活性較正常組顯著升高(P＜0.05)。與模型組相比,針刺組CSE活性明顯降低,且差異具有統(tǒng)計學意義(P＜0.05)；其結果說明針刺治療能降低結腸組織中CSE的活性。通過針刺“足三針”穴組,腹部術后大鼠結腸ICCs修復微環(huán)境中CSE活性顯著降低。研究結論針刺“足三針”穴組能明顯改善腹部術后大鼠的胃腸功能障礙,調整大鼠腹部術后結腸Cajal細胞修復微環(huán)境中CSE的活性,從而改善胃腸功能障礙的癥狀。
[Abstract]:Objective to observe the effect of acupuncture on the expression of CSE in the repair microenvironment of colon Cajal cells after abdominal operation in rats.
