不同腧穴配伍電針防治大鼠心肌缺血損傷作用的比較

【摘要】:目的:比較不同腧穴配伍電針對心肌缺血大鼠針效的差異,為臨床防治心肌缺血的選穴配伍提供依據(jù)。方法:Wistar雄性大鼠隨機分為正常組、模型組、標本配穴組(雙"內(nèi)關(guān)"配伍雙"足三里""關(guān)元")、常規(guī)配穴組(雙"內(nèi)關(guān)"配伍雙"神門""膻中"),每組各10只。鹽酸異丙腎上腺素(5mL/kg)皮下注射法復(fù)制心肌缺血模型。標本配穴組和常規(guī)配穴組在每日造模前電針處理10min,造模7d結(jié)束后仍每日電針,連續(xù)21d。檢測各組大鼠心電圖,采用全自動生化儀檢測血清肌鈣蛋白T(cTnT)水平,酶聯(lián)免疫法檢測心肌組織丙二醛(MDA)含量及超氧化物歧化酶(SOD)、谷胱甘肽過氧化物酶(GSH-Px)活性,透射電鏡觀察心肌細胞超微結(jié)構(gòu)變化。結(jié)果:與正常組比較,模型組心電圖S-T段振幅、心率(HR),血清cTnT和心肌組織MDA含量均顯著升高(P0.01),心肌組織SOD和GSH-Px活性顯著下降(P0.01),心肌超微結(jié)構(gòu)破壞明顯,線粒體腫脹變形,甚至空泡化;電針干預(yù)后,兩電針組心電圖S-T段振幅、HR,血清cTnT和心肌組織MDA含量顯著下降(P0.05),心肌組織SOD和GSH-Px活性顯著升高(P0.05),心肌超微結(jié)構(gòu)損傷減輕;兩電針組組間比較,標本配穴組血清cTnT和心肌組織MDA含量明顯低于常規(guī)配穴組(P0.05),心肌組織SOD和GSH-Px活性明顯高于常規(guī)配穴組(P0.05),標本配穴組心肌超微結(jié)構(gòu)損傷程度較常規(guī)配穴組輕。結(jié)論:電針能有效防治心肌缺血,標本配穴法電針的防治作用優(yōu)于常規(guī)配穴。
[Abstract]:Objective: to compare the acupuncture effect of different acupoints combined with electroacupuncture on myocardial ischemia rats. Methods: male Wistar rats were randomly divided into normal group and model group. The specimens were divided into two groups (double "Neiguan" and "Zusanli" and "Guanyuan") and conventional acupoint group (double "Neiguan" and "Shenmen" and "Shanzhong"). Myocardial ischemia model was established by subcutaneous injection of isoproterenol hydrochloride (5 mL / kg) in each group. The electrocardiogram (ECG) of the rats in each group was measured and the serum troponin TnTnT level was measured by automatic biochemical instrument after 7 days of modeling. The content of malondialdehyde (MDA) and the activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) in myocardium were detected by enzyme-linked immunosorbent assay (Elisa). Results: compared with the normal group, the amplitude of S-T segment and heart rate of the model group were compared with those of the normal group. The contents of serum cTnT and myocardial MDA increased significantly (P 0.01), and the activities of SOD and GSH-Px decreased significantly (P 0.01). The ultrastructure of myocardium was destroyed obviously. Mitochondria swelling and deformation, even vacuolation; After electroacupuncture intervention, the amplitudes of S-T segment of electrocardiogram (S-T) and the contents of serum cTnT and myocardial MDA in the two electroacupuncture groups decreased significantly (P 0.05). The activities of SOD and GSH-Px in myocardial tissue increased significantly (P 0.05), and the myocardial ultrastructure injury was alleviated. The contents of serum cTnT and myocardial MDA were significantly lower in group A than those in group B (P 0.05). The activities of SOD and GSH-Px in myocardial tissue were significantly higher than those in the control group (P0.05). Conclusion: electroacupuncture can effectively prevent and treat myocardial ischemia.
