【摘要】:第一部分古日古木-7是一種重要的傳統(tǒng)蒙藥復(fù)方,被廣泛應(yīng)用于肝病的治療。本研究探討了古日古木-7活性部位(大孔樹脂分離的甲醇洗脫部分,MF)對四氯化碳導(dǎo)致小鼠肝損傷的保護作用,并用超高效液質(zhì)聯(lián)用儀(UPLC-MS)分析了小鼠血清中的膽酸含量的變化以及MF中的活性成分。結(jié)果表明,連續(xù)給MF(66.132.264 mg/kg)四天,每天一次可以抑制四氯化碳(CCl4)誘導(dǎo)的小鼠肝臟組織病理學(xué)變化和血清轉(zhuǎn)氨酶的改變,并且具有一定的量效關(guān)系(組間比較用t檢驗(P0.05具顯著差異;P0.01具極顯著差異),結(jié)果發(fā)現(xiàn)丙氨酸轉(zhuǎn)氨酶(ALT)為2766.0±1909.7U/L、1929.0±799.8U/L.1144.0 ±787.2U/L(較模型組2461.8±1072.7U/L有顯著差異,P0.05);天冬氨酸轉(zhuǎn)氨酶(AST)為2482.8±1853.3U/L.1748.0±694.8U/L. 1173±785.3U/L(較模型組2506.6±1140.7U/L有極顯著差異,P0.01)。MF給藥組的超氧化物歧化酶(SOD)較四氯化碳組(75.75 ±5.37U/mg protein)有所增加:82.46±15.48U/mg protein.93.39± 40.91 U/mg protein.108.19±30.32U/mg protein(P0.01),但丙二醛(MDA)較四氯化碳組(44.32±16.68nmol/mg protein)水平有所降低:19.31 ± 6.58nmol/mg protein (P0.01).8.58±1.11 nmol/mg protein (P0.01).7.68±1.95nmol/mg protein (P0.01)。MF組和水飛薊素組的小鼠血清中,4種;撬峤Y(jié)合型膽汁酸含量明顯高于空白組。定量分析顯示,MF含有大量的鞣花酸(297.97mg/g)、木犀草素與其糖苷(35.10mg/g)、芹菜素與其糖苷(30mg/g)、熊果酸(14.91mg/g)、bidenoside C (8.75 mg/g)、原花青素(15.64mg/g的原花青素A2相當(dāng)量)。與古日古木-7相比,MF的有效量和化合物的數(shù)量都有所減少,為開發(fā)用量少并且易于質(zhì)量控制的新保肝藥物打下了基礎(chǔ)。第二部分鎖陽(Cynomorium songaricium)是一種無葉綠素,全寄生的植物,主要集中于我國的西北部。本研究以鎖陽為原料,制備鎖陽產(chǎn)品。將鎖陽粉碎成豌豆大小的顆粒,用95%的乙醇提取鎖陽中的三萜成分,以含有三萜成分的乙醇提取物制備鎖陽三萜咀嚼片,確定配方為:30%鎖陽三萜、15%奶粉、17%糖粉、27%玉米淀粉、10%微晶纖維素、0.22%甘草酸、0.78%檸檬酸等;將乙醇提取后的殘渣再用半胱氨酸加維生素C或者水在60℃下浸泡提取48 h,將所得殘渣清洗、烘干粉碎后過40目篩制得的兩種膳食纖維,測定兩種膳食纖維的物理性質(zhì),結(jié)果發(fā)現(xiàn),由半胱氨酸酸溶液處理的所得的膳食纖維的持水性為9.20 g水/g樣品,溶脹性為7.5 mL/g、水處理的膳食纖維這兩種性質(zhì)分別是5.83g水/g樣品;4.4 mL/g,與水處理的相比,半胱氨酸酸溶液處理后所得的膳食纖維質(zhì)量明顯較優(yōu)。
[Abstract]:The first part is an important traditional Mongolian medicine compound, which is widely used in the treatment of liver disease. The active site (methanol elution part of macroporous resin) was studied in this study. The protective effect of MF on liver injury induced by carbon tetrachloride in mice. The changes of serum cholic acid and the active components in MF were analyzed by UPLC-MS. MF(66.132.264 mg / kg for four days could inhibit the liver histopathological changes and serum aminotransferase changes induced by CCL _ 4). And there was a certain dose-effect relationship (significant difference was found between the two groups by t test (P0.05). The quantitative analysis showed that there was a large amount of tannic acid in the MF (297.97 mg / g). Luteolin and its glycoside 35.10 mg / g, apigenin and its glycoside 30 mg / g, ursolic acid 14.91 mg / g). Bidenoside C is 8.75 mg / g, and procyanidin is 15.64 mg / g, a significant amount of procyanidin A2. Both the effective amount of MF and the number of compounds decreased. It lays the foundation for the development of new hepatoprotective drugs with low dosage and easy quality control. The second part Cynomorium songaricium is a kind of chlorophyll free. The whole parasitic plant is mainly concentrated in the northwest of China. In this study, Chaoyang was used as the raw material to prepare the product of Chaoyang, which was crushed into pea-sized granules, and the triterpenoids were extracted by 95%% ethanol. Chaoyang triterpene chewable tablets were prepared from ethanol extract containing triterpenes, and the formula was determined as 10% microcrystalline cellulose.
