【摘要】：目的:探討針刺對脂多糖(lipopolysaccharide,LPS)誘導的慢性炎癥疼痛行為的影響以及對慢性炎癥的改善機制。方法:將24只C57BL/6J雄性小鼠(6-8周齡,22±5g)隨機分為3組:空白組小鼠腹腔注射4ml.kg-1生理鹽水;模型組小鼠腹腔注射脂多糖(LPS)3mg.kg-1,制造慢性炎癥小鼠模型;針刺組對于慢性炎癥小鼠給予針刺雙側"足三里"穴治療。對于三組小鼠不同處理后,分別采用Von Frey檢測各組小鼠的機械縮足反射閾值(mechanical withdrawal threshold,MWT),采用酶聯(lián)免疫(euzymelinked immunosorbent assay,ELISA)法檢測各組小鼠腫瘤壞死因子α(tumor necrosis factor,TNF-α)含量,采用蛋白質免疫印跡雜交(Western blot)法檢測各組小鼠背根神經節(jié)(dorsal root ganglion,DRG)神經元與香草酸瞬時受體亞型1(transient receptor potential vanilloid 1,TRPV 1)表達的差異,采用免疫組織化學法檢測各組小鼠DRG中TRPV1蛋白陽性細胞數(shù)的表達。結果:針刺組與模型組相比較,小鼠機械痛閾明顯升高(P0.05);血清TNF-α含量明顯降低(P0.05);DRG神經元TRPV1蛋白表達降低(P0.05),DRG神經元TRPV1蛋白陽性細胞數(shù)的表達降低(P0.05)。結論:針刺治療可以減少機體炎癥因子的表達,同時也改變DRG神經元中TRPV1的表達,從而緩解和抑制慢性炎癥所導致的疼痛。
[Abstract]:Objective: to study the effect of acupuncture on lipopolysaccharide of lipopolysaccharide. Methods: 24 C57BL / 6J male mice were selected for 6-8 weeks of age. 22 鹵5g) were randomly divided into three groups: mice in blank group were intraperitoneally injected with 4 ml 路kg ~ (-1) normal saline; The model group mice were injected intraperitoneally with lipopolysaccharide (LPSN) 3 mg 路kg ~ (-1) to produce chronic inflammatory mice model. Acupuncture group for chronic inflammation mice were given acupuncture bilateral "Zusanli" point treatment. For the three groups of mice after different treatment. Von Frey was used to detect the mechanical withdrawal threshold of mechanical reflex in each group. Enzyme linked immunized immunosorbent assay was used. The content of tumor necrosis factor- 偽 (TNF- 偽) in mice was determined by Elisa. The root ganglion of dorsal root ganglion (DRG) was detected by Western blot. Transient receptor potential vanilloid 1 between DRG neurons and vanillic acid transient receptor subtype 1. The expression of TRPV1 protein was detected by immunohistochemical method in DRG of mice in each group. Results: compared with model group, the expression of TRPV1 protein in acupuncture group was compared with that in model group. The mechanical pain threshold increased significantly in mice. The content of serum TNF- 偽 decreased significantly (P 0.05). The expression of TRPV1 protein in DRG neurons was decreased (P 0.05). The expression of TRPV1 protein positive cells in DRG neurons decreased P0.05.Conclusion: acupuncture treatment can reduce the expression of inflammatory factors. It also changes the expression of TRPV1 in DRG neurons, which alleviates and inhibits the pain caused by chronic inflammation.
