穿心蓮內(nèi)酯對病毒性心肌炎小鼠保護作用及其機制的研究

【摘要】:背景與目的病毒性心肌炎(Viral Myocarditis,VMC)是病毒侵犯心臟引起的以心肌細胞的變性壞死和心肌間質(zhì)炎癥為主的一種疾病。VMC的發(fā)病機制目前仍不十分清楚,治療上缺乏有效而特異的手段。本研究通過柯薩奇病毒B3(Coxsackievirus B3,CVB3)感染Balb/c小鼠,建立小鼠VMC模型,并采用穿心蓮內(nèi)酯注射液進行治療干預(yù),觀察穿心蓮內(nèi)酯對病毒性心肌炎小鼠模型的作用并探討其可能機制。方法1.150只雄性Balb/c小鼠隨機分為正常對照組(對照組,n=40),病毒感染組(感染組,n=60),穿心蓮內(nèi)酯治療組(治療組n=50)。2.感染組與治療組腹腔接種0.2ml含CVB3的病毒液,對照組注射等體積的生理鹽水。治療組0.5h后腹腔注射0.2ml 125mg/kg.d穿心蓮內(nèi)酯注射液,連續(xù)用藥2周,對照組與感染組每天注射等體積的生理鹽水。3.觀察小鼠生存狀態(tài),統(tǒng)計小鼠死亡率,于病毒接種后7d和14d觀察心肌組織病理學(xué)改變、血清白細胞介素6(IL-6)水平、心肌組織中腫瘤壞死因子α(TNF-α)表達水平。結(jié)果1.小鼠接種病毒第7天及14天,與對照組比較,感染組小鼠血清IL-6、心肌TNF-α表達、心肌病理評分顯著增加,差異有統(tǒng)計學(xué)意義(P0.05)。2.小鼠病毒接種后7天及14天,與感染組相比,治療組小鼠血清IL-6、心肌TNF-α表達、心肌病理評分降低,差異有統(tǒng)計學(xué)意義(P0.05)。結(jié)論穿心蓮內(nèi)酯能夠減輕病毒性心肌炎小鼠心肌損害。其機制可能抑制血清IL-6、心肌TNF-α表達水平,減輕心肌炎心肌損傷。
[Abstract]:Background and objective virus Myocarditis in viral myocarditis. VMC is a kind of disease caused by virus invading the heart. The pathogenesis of VMC is not clear at present. It is mainly caused by degeneration and necrosis of myocardial cells and interstitial inflammation of myocardium. In this study, Balb/c mice were infected with Coxsackievirus B3Coxsackievirus B3CVB3. Mouse VMC model was established and treated with andrographolide injection. To observe the effect of andrographolide on viral myocarditis in mice and explore its possible mechanism 1.150 male Balb/c mice were randomly divided into normal control group (control group 40). Virus infection group (infection group), andrographis lactone treatment group (treatment group) (nonglactide treatment group). The infection group and the treatment group were intraperitoneally inoculated with 0.2 ml of virus solution containing CVB3. The control group was injected with the same volume of normal saline, and the treatment group was injected with 0.2 ml 125mg / kg 路d andrographolide injection for 2 weeks after 0.5 h. Control group and infection group were injected with saline of the same volume every day. The survival status of mice was observed and the mortality of mice was counted. The pathological changes of myocardium were observed 7 and 14 days after inoculation. The level of serum interleukin-6 (IL-6) and the expression of TNF- 偽 in myocardial tissue. Results 1.The mice were inoculated with the virus on the 7th and 14th day, compared with the control group. Serum IL-6, TNF- 偽 expression and cardiomyopathology score were significantly increased in the infected group. The difference was statistically significant (P0.05. 2.The mice were inoculated 7 and 14 days after inoculation). Compared with the infection group, the serum IL-6, TNF- 偽 expression and cardiomyopathology score in the treatment group were lower than those in the control group. Conclusion andrographolide can reduce myocardial damage in mice with viral myocarditis and its mechanism may inhibit the expression of serum IL-6 and myocardial TNF- 偽. Reduce myocardial injury in myocarditis.
