電針膽下合穴陽陵泉等對AC模型豚鼠α7nAchR及TNF-α的影響

【摘要】:目的:對比觀察電針急性膽囊炎(AC)模型豚鼠陽陵泉、上巨虛、下巨虛、足三里穴對膽囊組織中腫瘤壞死因子-α(TNF-α)、煙堿型乙酰膽堿受體α7(α7nAchR)及血清中的高遷移率族蛋白B1(HMGB1)表達水平的影響,探討膽之下合穴陽陵泉治療膽腑病是否存在相對特異性。方法:將72只健康SD豚鼠隨機分為空白、模型、陽陵泉、上巨虛、下巨虛、足三里組共六組,每組12只,雌雄各半。除空白組外均膽囊注射大腸桿菌建立豚鼠急性膽囊炎模型,造模成功并治療5天后,取膽囊組織進行HE染色,觀察各組豚鼠膽囊形態(tài)學變化,運用Western blot法檢測豚鼠膽囊組織α7 nAchR受體的表達,免疫組化法檢測膽囊組織中TNF-α的表達,ELISA法檢測豚鼠血清中HMGB1水平。結果:1.光鏡下膽囊形態(tài)學的觀察(10x40倍),結果顯示:空白組:粘膜上皮細胞為單層柱狀上皮,排列整,固有膜較薄,含部分小血管和淋巴管,固有膜及粘膜下層無淤血擴張,基層排列不規(guī)整,漿膜完整,無炎癥反應。模型組:有大量粘膜上皮壞死脫落,充血水腫,固有層有大量炎性細胞浸潤。陽陵泉組:粘膜上皮大部分完好,固有層松弛,有充血水腫,部分淋巴細胞浸潤,有較少中性粒浸潤,肌層和漿膜層充血。其他穴位組:粘膜上皮有部分脫落,固有層及粘膜下層及肌層有中性粒細胞浸潤,血管明顯擴張淤血。2.各組豚鼠組織α7nAch R含量檢測,結果顯示:與空白組比,余組膽囊組織中α7nAchR表達水平均明顯偏低(P0.01),差異有顯著的統(tǒng)計學意義(P0.01)。電針治療后與模型組比較,各電針治療組膽囊組織中α7nAchR表達均明顯偏高(P0.01),差異有顯著的統(tǒng)計學意義(P0.01)。與陽陵泉比較,上巨虛、下巨虛、足三里組豚鼠膽囊組織中α7nAchR表達均偏低,差異有統(tǒng)計學意義(P0.01或P0.05)。3.各組豚鼠血清HMGB1含量檢測,結果顯示:與空白組比,血清中HMGB1含量表達均明顯偏高,差異有顯著的統(tǒng)計學意義(P0.01)。與模型組比較,各電針治療組血清中HMGB1含量表達均明顯偏低,差異有顯著的統(tǒng)計學意義(P0.01)。與陽陵泉比較,上巨虛、下巨虛、足三里組血清中HMGB1含量表達均偏高,差異有顯著的統(tǒng)計學意義(P0.01)。4.各組組織中TNF-α的表達檢測,結果顯示:與空白組比,膽囊組織中TNF-α表達均明顯偏高,差異有顯著的統(tǒng)計學意義(P0.01)。電針治療后與模型組比,膽囊組織中TNF-α表達均明顯偏低,差異有顯著的統(tǒng)計學意義(P0.01)。與陽陵泉組比,上巨虛組、下巨虛組、足三里組膽囊組織中TNF-α表達均偏高,差異有統(tǒng)計學意義(P0.01或P0.05)。結論1.電針消化系相關的內腑下合穴對AC豚鼠均可產生不同程度的治療作用,其機制可能是通過調節(jié)α7nAchR、TNF-α表達及HMGB1含量而達到抑制炎癥因子、減輕炎癥反應、保護AC模型豚鼠膽囊粘膜實現的2.對比可知電針陽陵泉對AC豚鼠炎癥干預效果優(yōu)于上巨虛、下巨虛及足三里穴,表明電針陽陵泉穴治療AC具有一定的相對特異性。
[Abstract]:Objective: to observe the effect of electroacupuncture (EA) on tumor necrosis factor- 偽 (TNF- 偽) in gallbladder tissue of guinea pig (Yanglingquan, Shangjuxu, Xijuxu, Zusanli). Effects of nicotinic acetylcholine receptor 偽 7 (偽 7 nAchR1) and high mobility group protein B1 HMGB1 in serum. Methods: 72 healthy Sprague-Dawley guinea pigs were randomly divided into six groups: blank model, Yanglingquan, Shangjuxu, Xijuxu and Zusanli groups. The guinea pig model of acute cholecystitis was established by injecting Escherichia coli into the gallbladder except the blank group. The model was successfully established and treated for 5 days. The gallbladder tissue was stained with HE. The changes of gallbladder morphology in each group were observed, and the expression of 偽 7 nAchR receptor in gallbladder tissue of guinea pigs was detected by Western blot method. The expression of TNF- 偽 in gallbladder tissue was detected by immunohistochemical method and HMGB1 level in serum of guinea pigs was detected by Elisa. Results: the morphology of gallbladder was observed 10 x 40 times under light microscope). The results showed that in the blank group, the mucosal epithelial cells were monolayer columnar epithelium, arranged neatly, thin lamina propria, including some small blood vessels and lymphatic vessels, the lamina propria and submucous layer had no congestion and dilatation, and the basic layer was irregular. Model group: a large number of mucosal epithelial necrosis, hyperemia and edema, a large number of inflammatory cells infiltration in the lamina propria. Yanglingquan group: most of the mucosal epithelium intact, lamina propria relaxation. There were hyperemia and edema, some lymphocytic infiltration, less neutrophil infiltration, hyperemia in myometrium and serous layer. Other acupoints group: mucosal epithelium had partial exfoliation, neutrophil infiltration in lamina propria and submucous layer and myometrium. The content of 偽 7nAch R in guinea pig tissues of each group was detected. The results showed that compared with the blank group, the content of 偽 7nAch R was higher than that of the control group. The expression of 偽 7nAchR in gallbladder tissues in the remaining group was significantly lower than that in the model group (P 0.01), and the difference was statistically significant (P 0.01). After electroacupuncture treatment, the expression of 偽 7nAchR was compared with that of the model group. The expression of 偽 7nAchR in gallbladder tissues of each electroacupuncture treatment group was significantly higher than that of P0.01a, and the difference was statistically significant (P 0.01). Compared with Yangling Quan, the expression of 偽 7nAchR in the gallbladder tissue was significantly higher than that in the upper giant asthenia group and the lower giant asthenia group. The expression of 偽 7nAchR in gallbladder tissue of Zusanli group was lower than that of Zusanli group, and the difference was significant (P0.01 or P0.05). The serum HMGB1 content of each group was detected. The results showed that the expression of HMGB1 in serum was significantly higher than that in the blank group, and the difference was statistically significant (P 0.01), compared with the model group. The expression of HMGB1 in serum of each electroacupuncture treatment group was significantly lower than that of Yanglingquan group, and the difference was statistically significant (P 0.01). The expression of TNF- 偽 in serum of Zusanli group was higher than that of control group, and the difference was statistically significant (P 0.01). The results showed that the expression of TNF- 偽 was higher than that of blank group. The expression of TNF- 偽 in gallbladder tissue was significantly higher than that in the model group, and the difference was statistically significant (P 0.01). Compared with the model group, the expression of TNF- 偽 in gallbladder tissue was significantly lower than that in the model group. Compared with Yanglingquan group, the expression of TNF- 偽 in gallbladder tissues of upper giant asthenia group, lower giant asthenia group and Zusanli group was higher than that in Yanglingquan group. The difference is statistically significant P0.01 or P0.050.Conclusion 1. Electroacupuncture of the digestive system related to the internal fu-fu points can produce different degrees of treatment for AC guinea pigs. 2. The mechanism may be to inhibit the inflammatory factors and alleviate the inflammatory response by regulating the expression of 偽 7nAchR- 偽 TNF- 偽 and the content of HMGB1. The effect of electroacupuncture on inflammation of AC guinea pigs was better than that of upper giant asthenia, lower giant asthenia and Zusanli point. The results showed that the treatment of AC by electroacupuncture at Yanglingquan point had certain relative specificity.
