額爾敦—烏日勒對動脈粥樣硬化易損斑塊家兔血管內(nèi)皮功能的影響

【摘要】:目的通過觀測額爾敦-烏日勒對家兔動脈粥樣硬化易損斑塊模型血管內(nèi)皮因子ET、PAF、TXA-2、PGI2、VEGF、E-selectin、SDF-1以及血管內(nèi)皮凝血纖溶物質(zhì)t-PA、PAI-1、Fbg的影響,明確其穩(wěn)定斑塊的作用機制。方法將50只家兔,隨機分為正常組和造模組。正常組10只不進行造模,普通飼料喂養(yǎng);造模組40只采用高脂飲食喂養(yǎng)、免疫損傷、球囊拉傷術(shù)復(fù)合因素造模,建立家兔動脈粥樣硬化易損斑塊模型。8周末造模成功后,將造模組隨機分為模型組、辛伐他汀組、額爾敦-烏日勒組,正常組和模型組分別灌服同體積的蒸餾水,給藥容量均為4ml·kg-1·d-1;辛伐他汀組給予辛伐他汀5mg·kg-1·d-1;額爾敦-烏日勒組給予額爾敦-烏日勒0.4g·kg-1·d-1,連續(xù)給藥16周。第24周末實驗結(jié)束,將最終完成實驗的39只家兔(正常組10只、模型組10只、額爾敦-烏日勒組10只、辛伐他汀組9只)處死取材。觀測以下指標(biāo):1.運用免疫組化法檢測各組家兔主動脈血管內(nèi)皮因子ET、PAF、VEGF、SDF-1、t-PA、PAI的表達情況。2.運用Elisa方法檢測各組家兔血清中血管內(nèi)皮因子ET、PAF、TXA-2、PGI2、VEGF、E-selectin、SDF-1、t-PA、PAI、Fbg的濃度。結(jié)果1.免疫組化結(jié)果顯示:(1)額爾敦-烏日勒能降低主動脈ET、PAF表達(P0.01),在降低ET表達上優(yōu)于辛伐他汀(P0.05)。(2)與模型組相比,額爾敦-烏日勒組主動脈VEGF和SDF-1的表達顯著減少(P0.05),但對E-selectin的影響無顯著性差異(P0.05)。(3)與模型組比較,額爾敦-烏日勒組主動脈PAI-1的表達顯著降低(P0.01),且表達量低于辛伐他汀組(P0.05),但對t-PA的影響無統(tǒng)計學(xué)差異(P0.05)。2.Elisa檢測結(jié)果顯示:(1)與模型組比較,額爾敦-烏日勒顯著降低血清ET、PAF(P0.01),在降低ET水平上優(yōu)于辛伐他汀組。(2)額爾敦-烏日勒能顯著降低血清TXA-2的水平、升高PGI2的水平(P0.05)。(3)額爾敦-烏日勒能顯著降低家兔血清VEGF和SDF-1的濃度(P0.01),且對其降低SDF-1的濃度優(yōu)于辛伐他汀組(P0.05)。(4)與模型組比較,額爾敦-烏日勒組家兔血清PAI-1及Fbg水平顯著降低(P0.01),但對t-PA的影響無統(tǒng)計學(xué)差異(P0.05);結(jié)論額爾敦-烏日勒具有改善動脈粥樣硬化血管內(nèi)皮功能的作用,其可能通過降低血管內(nèi)皮因子ET、PAF、TXA-2、VEGF、SDF-1的水平以及升高血清PGI2的水平;同時也可通過降低血清血管內(nèi)皮凝血纖溶物質(zhì)PAI-1、Fbg水平來發(fā)揮抗動脈粥樣硬化和穩(wěn)定易損斑塊的作用。
[Abstract]:Objective to observe the effect of Erden-Uzhler on vascular endothelial factor (ETF) TXA-2 PGI2 (VEGFN) E-selectin in atherosclerotic vulnerable plaque model in rabbits. The effects of SDF-1 and the fibrinolytic substance t-PAI-1Fbg on the stability of plaque were determined. Methods 50 rabbits were selected. They were randomly divided into normal group and model group. The model group (40 rats) were fed with high fat diet, immune injury and balloon wound were used to make the model. The model group was randomly divided into model group after the successful establishment of atherosclerotic plaque model at the end of week .8. Simvastatin group, Erden-Uril group, normal group and model group were given distilled water of the same volume, respectively, and the dosage was 4ml 路kg-1 路d-1; Simvastatin group was given simvastatin 5 mg 路kg-1 路d -1; Erden-Uzhler group was given 0.4 g 路kg-1 路d-1 of Erden-Ujil for 16 weeks. At the end of the 24th week, 39 rabbits (10 normal rabbits) were selected to complete the experiment at the end of the 24th week. Ten rats in the model group, 10 in the Erden-Ure group and 9 in the simvastatin group were killed. The following indexes were observed: 1. The endothelium factor ETF-PAF in aorta of rabbits in each group was detected by immunohistochemical method. The expression of VEGFF-1t-PAI in rabbit serum was detected by Elisa method. Results 1.Immunohistochemical results showed that Erden-1) Erden-Uzhler could decrease the et of aorta. The expression of PAF was higher than that of simvastatin (P0.05, P0.05, P < 0.05), which was better than that of the model group in decreasing the expression of et. The expression of VEGF and SDF-1 in the aorta of Erden-Ujil group was significantly decreased (P 0.05). However, there was no significant difference in the effect of E-selectin on the expression of PAI-1 in the aorta of the Erden-Ujil group (P 0.05) compared with the model group (P 0.01). The expression of t-PA in simvastatin group was lower than that in simvastatin group, but there was no significant difference in the effect of t-PA. Erden-Uzhler significantly reduced the level of serum ETF-PAFN P0.01, which was superior to that of simvastatin group in lowering the level of TXA-2.) Erden-Uriel could significantly reduce the level of serum TXA-2. Raising the level of PGI2 (P0.05N.) Erden-Uzhler could significantly decrease the concentration of VEGF and SDF-1 in serum of rabbits (P 0.01). The decrease of SDF-1 concentration was better than that of simvastatin group (P0.05A. 4) and compared with the model group. The levels of serum PAI-1 and Fbg in Erden-Uzhler group decreased significantly (P 0.01), but there was no significant difference in the effect of t-PA on serum PAI-1 and Fbg. Conclusion Erden-Uzhler can improve the endothelial function of atherosclerotic vessels, which may be through the reduction of vascular endothelial factor ETF-PAFTXA-2VEGF.Conclusion Erden-Uzhler can improve the endothelial function of atherosclerotic vessels. The level of SDF-1 and the level of serum PGI2; At the same time, it can also play the role of anti-atherosclerosis and stabilization of vulnerable plaque by reducing the level of PAI-1 FBG.
