大黃素、茶多酚和魚腥草素鈉對(duì)TNBS誘導(dǎo)大鼠結(jié)腸炎的治療作用

【摘要】:目的:觀察大黃素、茶多酚和魚腥草素鈉對(duì)2,4,6-三硝基苯磺酸(TNBS)誘導(dǎo)大鼠結(jié)腸炎的治療作用,并探討其可能的免疫機(jī)制。方法:72只SPF級(jí)S-D大鼠隨機(jī)分為九組,為空白對(duì)照組(A組)、大黃素高劑量組(B組,50mg/kg)、大黃素低劑量組(C組,25mg/kg)、茶多酚高劑量組(D組,340mg/kg)、茶多酚低劑量組(E組,170mg/kg)、魚腥草素鈉高劑量組(F組,100mg/kg)、魚腥草素鈉低劑量組(G組,50mg/kg)、柳氮磺吡啶組(H組,100mg/kg)和模型組(I組)。造模當(dāng)日,B-I組大鼠均以75mg/kgTNBS乙醇溶液灌腸,A組以同體積30%乙醇溶液灌腸。造模24h后,B-H組定時(shí)以給定劑量的藥物灌胃,A組和I組以相同體積的蒸餾水灌胃。每天1次,持續(xù)14d。觀察大鼠每日體重變化、大便狀態(tài),解剖后觀察結(jié)腸組織大體改變和HE染色后病理變化并評(píng)分,檢測(cè)血清TNF-α、IL-lβ、IL-4、IL-10水平及ALT、AST和肌酐含量。結(jié)果:⑴與A組相比,B-H組和I組大鼠一般情況較差,體重明顯減輕,大體評(píng)分和組織病理評(píng)分均較低,TNF-α和IL-lβ水平明顯增高,IL-4和IL-10的表達(dá)顯著下降,差異有統(tǒng)計(jì)學(xué)意義(P0.05),但G組與I組相比,大部分指標(biāo)P0.05,差異無統(tǒng)計(jì)學(xué)意義。⑵與I組相比,B-H組一般情況較好,體重有所回升,大體狀況和組織病理顯示炎癥程度明顯減輕,且TNF-α和IL-lβ表達(dá)顯著下降,IL-4和IL-10的水平有明顯提升,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。⑶B-H組之間兩兩比較,B-F組所有指標(biāo)和H組相比以及B、C組兩組相比有差異均無統(tǒng)計(jì)學(xué)意義(P0.05);然而D、E兩組和F、G兩組分別比較P值均小于0.05,差異有統(tǒng)計(jì)學(xué)意義。⑷藥物肝腎毒性檢測(cè),B-I組大鼠ALT、AST值明顯高于A組,B-H組ALT、AST值明顯低于I組(P0.05);但A-I組肌酐值無顯著性差異(P0.05)。結(jié)論:⑴采用75mg/kg濃度的TNBS溶液誘導(dǎo),成功建立了大鼠結(jié)腸炎模型。⑵25mg/kg、50mg/kg劑量的大黃素,170mg/kg、340mg/kg的茶多酚和100mg/kg的魚腥草素鈉灌胃可減輕大鼠結(jié)腸病理組織炎癥和損傷,降低血清促炎因子TNF-α和IL-lβ水平,升高抑炎因子IL-4和IL-10水平,對(duì)大鼠結(jié)腸炎有治療作用,且未見肝腎毒性。⑶170mg/kg、340mg/kg劑量的茶多酚對(duì)大鼠結(jié)腸炎的治療作用存在量-效關(guān)系;而大黃素在50mg/kg、25mg/kg兩種劑量灌胃的情況下,對(duì)大鼠結(jié)腸炎治療作用無明顯差異。
[Abstract]:Objective: to observe the therapeutic effect of emodin, tea polyphenol and houttuynin sodium on rat colitis induced by TNBS. Methods Seventy-two SPF S-D rats were randomly divided into nine groups: blank control group A group and emodin high dose group B group (50 mg / kg). The emodin concentration was 25 mg / kg in C group, 340 mg / kg in D group and 170 mg / kg in E group (P < 0.05). High dose of houttuyfonate sodium group (100 mg / kg), low dose group (50 mg / kg) of sodium houttuynate sodium and sulfasalopyridine group (H group). On the same day, the rats in the B-I group were enema with 75 mg / kg TNBS ethanol solution. The rats in group A were perfused with the same volume of 30% ethanol solution 24 hours later, the rats in group B and H were perfused with the same volume of distilled water once a day. For 14 days, the changes of body weight, stool status, colonic gross changes and pathological changes after HE staining were observed, and the serum TNF- 偽 IL-l 尾 and IL-4 were detected.
