CXCR4/CXCL12生物軸促進(jìn)結(jié)腸癌細(xì)胞侵襲轉(zhuǎn)移作用的研究

研究背景結(jié)直腸癌(colorectal cancer, CRC)是常見(jiàn)的消化道惡性腫瘤之一,其發(fā)病率和死亡率分別是第二位和第三位,嚴(yán)重威脅人類(lèi)的健康。結(jié)腸癌患者死亡的主要原因是轉(zhuǎn)移和局部復(fù)發(fā),為此人們對(duì)結(jié)腸癌轉(zhuǎn)移的機(jī)制進(jìn)行了大量的研究。腫瘤細(xì)胞轉(zhuǎn)移的步驟主要包括,腫瘤細(xì)胞原發(fā)灶過(guò)度增殖及血管生成,分泌膠原酶降解細(xì)胞外基質(zhì)(extracellular matrix, ECM)和基底膜,細(xì)胞從原發(fā)部位分離脫落,浸潤(rùn)血管或淋巴管并在血流中形成癌栓隨血流遷移,腫瘤細(xì)胞遷移至靶器官通過(guò)增殖和血管生成形成新生灶。腫瘤細(xì)胞的轉(zhuǎn)移是一個(gè)十分復(fù)雜的動(dòng)態(tài)的生物學(xué)過(guò)程,由多個(gè)相對(duì)獨(dú)立的步驟組成,每一個(gè)步驟涉及到多種分子事件。目前對(duì)于腫瘤細(xì)胞侵襲轉(zhuǎn)移機(jī)制的解釋很多,其中腫瘤“歸巢”學(xué)說(shuō)認(rèn)為某些腫瘤細(xì)胞高表達(dá)趨化因子受體,而靶器官高表達(dá)趨化因子,腫瘤細(xì)胞可以像白細(xì)胞一樣在趨化因子的誘導(dǎo)下向靶器官遷移。CXCL12是一種細(xì)胞趨化因子,其受體為細(xì)胞表面的G蛋白偶聯(lián)受體CXCR4,CXCL12與CXCR4相互作用形成偶聯(lián)分子對(duì),共同促進(jìn)白細(xì)胞和造血干細(xì)胞的遷移。近年來(lái)研究發(fā)現(xiàn),結(jié)腸癌組織中高表達(dá)CXCR4,而其最常見(jiàn)...
