轉(zhuǎn)化生長因子β1通過Wnt/β-catenin信號通路誘導(dǎo)人肝癌細(xì)胞SMMC-7721上皮間質(zhì)轉(zhuǎn)化的實(shí)驗(yàn)研究

肝細(xì)胞癌(Hepatocellular carcinoma, HCC)是目前我國臨床上較為常見的惡性腫瘤,位于常見腫瘤的第六位及腫瘤相關(guān)性死亡原因的第三位,且近年來呈現(xiàn)逐漸上升的趨勢。雖然隨著科學(xué)技術(shù)的不斷發(fā)展和外科手術(shù)技術(shù)的不斷提高,目前大部分早期確診的肝癌患者都能夠?qū)嵭懈涡郧谐?但是肝癌患者的5年總體生存率仍然很低。其中最重要的因素是肝癌具有極強(qiáng)的侵襲和轉(zhuǎn)移能力,術(shù)后5年內(nèi)極易出現(xiàn)腫瘤復(fù)發(fā)及遠(yuǎn)處轉(zhuǎn)移,很多患者在術(shù)后1-2年就會出現(xiàn)肝內(nèi)或肝外復(fù)發(fā)、轉(zhuǎn)移。所以,如何有效的控制肝癌術(shù)后復(fù)發(fā)轉(zhuǎn)移是我們治療肝癌的臨床難題。腫瘤微環(huán)境包括腫瘤細(xì)胞或者間質(zhì)細(xì)胞分泌的多種細(xì)胞因子、趨化因子和生長因子,它們共同組成了腫瘤細(xì)胞在體內(nèi)生長、增殖、甚至是轉(zhuǎn)移相關(guān)所必須的體內(nèi)微環(huán)境,在腫瘤發(fā)生發(fā)展和促進(jìn)腫瘤細(xì)胞侵襲轉(zhuǎn)移中都發(fā)揮著關(guān)鍵作用。轉(zhuǎn)化生長因子p1(Transforming growth factor-β1, TGF-β1)在肝臟等消化系統(tǒng)惡性腫瘤中發(fā)揮著復(fù)雜的調(diào)控作用,包括維持腫瘤干細(xì)胞的穩(wěn)態(tài),促進(jìn)肝纖維化進(jìn)程,作為腫瘤抑制因子參與免疫調(diào)節(jié)等。上皮間質(zhì)轉(zhuǎn)化(Epithelial-mesenchym...
