去泛素化酶USP42在調(diào)節(jié)胃癌KLF4蛋白降解中的作用

[研究背景]胃癌是嚴(yán)重影響我國及世界人民生命健康的惡性腫瘤。胃癌的發(fā)病原因尚不完全明確,它的發(fā)生發(fā)展是一個多因素多步驟的過程,幽門螺桿菌感染是其中的關(guān)鍵風(fēng)險因素。有文獻(xiàn)表明CagA在幽門螺桿菌誘導(dǎo)的胃癌中發(fā)揮著細(xì)菌癌蛋白的作用。在胃上皮細(xì)胞中,CagA能激活致癌基因的表達(dá),還能通過啟動子高度甲基化導(dǎo)致抑癌基因功能的失活,從而導(dǎo)致癌癥發(fā)生。CagA還可以通過腫瘤相關(guān)巨噬細(xì)胞來抑制免疫微環(huán)境,來促進(jìn)癌癥的發(fā)展。KLF4是一個含有鋅指結(jié)構(gòu)的轉(zhuǎn)錄因子,在終末分化的胃腸粘膜上皮細(xì)胞中高表達(dá),與腫瘤的發(fā)生發(fā)展有著密不可分的關(guān)系。還有一個位于轉(zhuǎn)錄激活結(jié)構(gòu)域和轉(zhuǎn)錄抑制結(jié)構(gòu)域的PEST序列,提示KLF4可能通過泛素-蛋白酶體通路降解。KLF4的半衰期短。它是公認(rèn)的胃腫瘤抑制基因。泛素化蛋白酶體途徑是調(diào)節(jié)細(xì)胞內(nèi)蛋白質(zhì)功能和降解的重要系統(tǒng),主要調(diào)節(jié)細(xì)胞內(nèi)一些結(jié)構(gòu)異;虬胨テ诙痰牡鞍。長期以來泛素化的功能研究專注于泛素化酶系統(tǒng)。然而,近些年的研究表明,去泛素化酶(DUBs)也積極參與癌癥中的細(xì)胞事件,我們課題組所關(guān)注的DUBs的功能是通過加工泛素基因產(chǎn)物,重塑多泛素鏈,逆轉(zhuǎn)泛素添加到靶蛋白的過程。與其他酶促蛋...
