CARD10通過(guò)調(diào)節(jié)NF-κB信號(hào)通路促進(jìn)腎癌進(jìn)展

【研究背景】腎細(xì)胞癌(簡(jiǎn)稱(chēng)腎癌)是泌尿科常見(jiàn)的腫瘤,近年來(lái)每年新發(fā)病例數(shù)一直顯現(xiàn)逐年上升的情形。腎癌的早期癥狀不明顯,診斷時(shí)約1/4-1/3病人已發(fā)生轉(zhuǎn)移。發(fā)生轉(zhuǎn)移的腎癌往往失去手術(shù)機(jī)會(huì),但發(fā)展至此階段的病人藥物治療效果不佳,因此需要進(jìn)一步研究腎癌的發(fā)生機(jī)制,為治療提供新的方向。CARD10(半胱天冬酶募集結(jié)構(gòu)蛋白10)又叫做CARMA3,已經(jīng)有研究表明CARD10的表達(dá)與腫瘤的發(fā)生密切相關(guān),其作用主要是通過(guò)激活轉(zhuǎn)錄因子NF-κB促進(jìn)腫瘤進(jìn)展。然而,此信號(hào)通路在腎癌中的是否存在仍不清楚!狙芯磕康摹緾ARD10是否在腎癌細(xì)胞中高表達(dá),抑制CARD10表達(dá)是否能抑制腎癌細(xì)胞的增殖、侵襲、遷移、上皮間質(zhì)轉(zhuǎn)化和促進(jìn)其凋亡,及其能否通過(guò)NF-κB信號(hào)通路進(jìn)行調(diào)節(jié)?【研究方法】1.首先培養(yǎng)ACHN細(xì)胞、786-O細(xì)胞和HK-2細(xì)胞,并提取其總蛋白,使用蛋白免疫印跡法檢測(cè)CARD10在三種細(xì)胞的表達(dá)情況。2.然后用sh RNA進(jìn)行細(xì)胞轉(zhuǎn)染腎癌ACHN和786-O細(xì)胞,形成不含目的基因(sh GFP)轉(zhuǎn)染的陰性對(duì)照組和含目的基因(特異性CARD10sh RNA)轉(zhuǎn)染的實(shí)驗(yàn)組細(xì)胞,在蛋白分子水平檢測(cè)其...
