RAGE和DIAPH1對(duì)肺腺癌細(xì)胞遷移和凋亡的影響及其在非小細(xì)胞肺癌組織中的表達(dá)

研究背景肺癌是造成全世界發(fā)病率和死亡率較高的主要原因之一,目前已成為全世界死亡率最高的惡性腫瘤,嚴(yán)重危害人類的生命健康。正因?yàn)榉伟┰谄湓缙陔A段對(duì)周圍血管造成的浸潤(rùn),進(jìn)一步導(dǎo)致其發(fā)生遠(yuǎn)處轉(zhuǎn)移,使得失去了手術(shù)治療的機(jī)會(huì)。因此,尋找控制肺癌的浸潤(rùn)轉(zhuǎn)移的有效治療靶點(diǎn)變得極為重要。目的1、研究晚期糖基化終產(chǎn)物受體(receptor for advanced glycation end products,RAGE)及其胞內(nèi)信號(hào)分子DIAPH1在肺腺癌細(xì)胞株A549中的表達(dá)及對(duì)細(xì)胞遷移及凋亡能力的影響。2、觀察RAGE及DIAPH1在非小細(xì)胞肺癌組織標(biāo)本中的表達(dá)情況,了解其在肺癌中的臨床價(jià)值。方法1、以肺腺癌細(xì)胞株A549及正常人支氣管上皮細(xì)胞BEAS-2B為研究對(duì)象,利用q-RT PCR、Western Blot及免疫細(xì)胞熒光染色,檢測(cè)RAGE及DIAPH1在二者中的表達(dá)及分布;2、RAGE配體CML-AGE及S100B以不同濃度(1μg、10μg、100μg)處理肺腺癌細(xì)胞A549,劃痕實(shí)驗(yàn)檢測(cè)對(duì)A549細(xì)胞遷移能力的影響;3、RAGE配體CML-AGE以不同濃度(25μg、50μg、100μg...

(A) RAGE andDIAPH1 expression in A549 cells; (B) RAGE and DIAPH1 expression in BEAS-2B cells.3.4 RAGE 配體 CML-AGE 及 S100B 對(duì)肺腺癌細(xì)胞株 A549 細(xì)胞遷移能力的影響劃痕實(shí)驗(yàn)結(jié)果顯示,以不同濃度 RAGE 配體 CML-AGE 及 S100B 刺激 A549肺腺癌細(xì)胞株。16h 后,相比于空白對(duì)照組,CML-AGE 實(shí)驗(yàn)組及 S100B 對(duì)細(xì)胞遷移能力有明顯抑制作用,其作用呈現(xiàn)出濃度依賴性。組與組之間使用 one-wayANOVA 進(jìn)行檢驗(yàn),差異有統(tǒng)計(jì)學(xué)意義 。(P<0.05)。見圖 4
