自發(fā)性乳腺腫瘤C3H/HeJNju小鼠原發(fā)腫瘤組織與脾臟間CD4 + CD25 + Treg TCR β CDR3組庫(kù)異

目的:采用Illumina Solexa高通量測(cè)序技術(shù),分析自發(fā)性乳腺腫瘤C3H/HeJNju小鼠原發(fā)乳腺腫瘤組織與脾臟間CD4+CD25+TregTCRβ CDR3組庫(kù)的同質(zhì)性與異質(zhì)性,探討腫瘤組織與和全身免疫循環(huán)之間CD4+CD25+Treg的區(qū)別和聯(lián)系,為進(jìn)一步研究乳腺癌中Treg的機(jī)制與應(yīng)用提供基礎(chǔ)。方法:1.10月齡以上的自發(fā)性乳腺腫瘤模型C3H/HeJNju出瘤小鼠3只,分別標(biāo)記為M1、M2、M3。2.分別采集M1、M2、M3小鼠原發(fā)乳腺腫瘤組織(T)和脾臟(S),采用磁珠分選技術(shù)分選出各組織CD4+CD25+Treg細(xì)胞,FCM鑒定純度。3.提取各分選獲得的CD4+CD25+Treg細(xì)胞基因組DNA,樣本序號(hào)命名:M1T:M1小鼠乳腺腫瘤組織樣本;M1S:M1小鼠脾臟樣本;M2T:M2小鼠乳腺腫瘤組織樣本;M2S:M2小鼠脾臟樣本;M3T:M3小鼠乳腺腫瘤組織樣本;M3S:M3小鼠脾臟樣本。4.基因組DNA樣本送Adaptive Biotechnologies Immuno SEQ公司(美國(guó)華盛頓醫(yī)學(xué)院),經(jīng)檢測(cè)符合建庫(kù)要求后,按小鼠TCR β CDR3組庫(kù)制備技術(shù)建立每...
