miRNA-199a-3p調(diào)控MAP3K4在胃癌中的表達(dá)

【摘要】:目的探討胃癌中MIRNA-199A-3P調(diào)控候選基因MAP3K4的表達(dá)。方法收集35例胃癌及癌旁組織標(biāo)本,采用莖環(huán)逆轉(zhuǎn)錄-聚合酶鏈?zhǔn)椒磻?yīng)法檢測(cè)胃癌組織及癌旁組織中MIR-199A-3P的表達(dá)水平,WESTERN BLOT法檢測(cè)MAP3K4表達(dá);細(xì)胞轉(zhuǎn)染法檢測(cè)MIR-199A-3P調(diào)控MAP3K4表達(dá),雙熒光素酶報(bào)告基因檢測(cè)系統(tǒng)驗(yàn)證MIR-199A-3P結(jié)合MAP3K4的3’UTR的靶位點(diǎn)。結(jié)果與癌旁組織相比,胃癌組織MIR-199A-3P高表達(dá),且其候選基因MAP3K4低表達(dá);MIR-199A-3P MIMICS轉(zhuǎn)染抑制MAP3K4表達(dá);MAP3K4是MIR-199A-3P作用的靶基因。結(jié)論 MIR-199A-3P是胃癌的一個(gè)癌基因,MAP3K4是受其調(diào)控的靶基因。
[Abstract]:Objective to investigate the expression of MIRNA-199A-3P regulatory candidate gene MAP3K4 in gastric cancer. Methods 35 specimens of gastric cancer and adjacent tissues were collected. The expression of MIR-199A-3P in gastric cancer and adjacent tissues was detected by, WESTERN BLOT method. The expression of MAP3K4 in gastric cancer tissues and adjacent tissues was detected by reverse transcription-polymerase chain reaction (RT-PCR). Cell transfection assay was used to detect the expression of MAP3K4 regulated by MIR-199A-3P and double luciferase reporter gene detection system was used to verify the target site of 3'UTR binding to MAP3K4 by MIR-199A-3P. Results compared with the paracancerous tissues, MIR-199A-3P was highly expressed and its candidate gene MAP3K4 was low; MIR-199A-3P MIMICS transfection inhibited the expression of MAP3K4; MAP3K4 was the target gene of MIR-199A-3P. Conclusion MIR-199A-3P is a oncogene of gastric cancer, and MAP3K4 is a target gene regulated by it.
