MicroRNA-27a在青海藏族胃癌組織中的表達(dá)和臨床病理關(guān)系的相關(guān)分析

【摘要】:目的:檢測(cè)青海地區(qū)藏族胃癌組織與癌周邊組織中miR-27a的表達(dá)水平,比較miR-27a在兩者不同組織中的表達(dá)水平是否具有差異;分析胃癌組織中miR-27a的相對(duì)定量和臨床相關(guān)病理資料是否具有相關(guān)性;分析從而探討miR-27a在青海藏族胃癌組織中的表達(dá)意義;并與其他民族胃癌組織中的表達(dá)情況進(jìn)行對(duì)比分析。方法:收集藏族胃癌病理組織標(biāo)本50例,分別將胃癌組織及癌旁組織取材并包埋成病理組織石蠟蠟塊,從石蠟蠟塊標(biāo)本內(nèi)提取miRNA、進(jìn)行逆轉(zhuǎn)錄并采取RT-PCR方法檢測(cè)胃癌組織和癌周邊組織兩者中mi R-27a的相對(duì)定量。并同時(shí)收集匯總藏族胃癌患者的臨床相關(guān)病理資料,并結(jié)合檢測(cè)結(jié)果進(jìn)行統(tǒng)計(jì)分析。檢索其他民族相關(guān)文獻(xiàn)中關(guān)于胃癌組織中miR-27a的報(bào)道,進(jìn)行對(duì)比探討。結(jié)果:(1)MiR-27a在藏族胃癌組織中的表達(dá)水平為7.415±0.129,在藏族癌旁組織的表達(dá)水平為2.050±0.062,兩組之間的表達(dá)差異具有統(tǒng)計(jì)學(xué)的意義(P0.05),即miR-27a在青海藏族胃癌組織中的表達(dá)高于其癌旁組織。(2)Mi R-27a在淋巴結(jié)發(fā)生癌轉(zhuǎn)移的胃癌組織中的表達(dá)水平高于未發(fā)生淋巴結(jié)癌轉(zhuǎn)移者,兩者間的差異具有統(tǒng)計(jì)學(xué)意義(P0.05),而miR-27a的表達(dá)水平與其他臨床相關(guān)的病理因素如診斷時(shí)年齡、患者的性別及其胃癌腫瘤組織分化程度無顯著相關(guān)性。(3)早期胃癌患者(Ⅰ-Ⅱ期)與晚期胃癌患者(Ⅲ-Ⅳ期)的癌組織中miR-27a的表達(dá)水平未見明顯差異。結(jié)論:(1)藏族胃癌組織中miR-27a的表達(dá)水平上調(diào),并且其相對(duì)定量與淋巴結(jié)是否癌轉(zhuǎn)移具有一定相關(guān)性,提示miR-27a對(duì)疾病的診斷有一定提示意義,并可推測(cè)胃癌的進(jìn)展及預(yù)后情況。(2)藏族胃癌早期與晚期患者中mi R-27a的表達(dá)量未見明顯差異,推測(cè)miR-27a可能參與胃癌早期啟動(dòng)的發(fā)生過程。(3)藏族胃癌組織中miR-27a的表達(dá)情況與其他民族文獻(xiàn)記錄相一致。
[Abstract]:Objective: to detect the expression level of miR-27a in Tibetan gastric cancer tissues and surrounding tissues in Qinghai, and to compare the expression level of miR-27a in different tissues. To analyze the correlation between the relative quantitative and clinicopathological data of miR-27a in gastric cancer tissues, and to explore the significance of miR-27a expression in Tibetan gastric cancer tissues of Qinghai province. The expression of gastric cancer in other nationalities was compared with that in other ethnic groups. Methods: 50 specimens of gastric cancer were collected, and the samples of gastric cancer tissues and adjacent tissues were collected and embedded into paraffin wax blocks of pathological tissues, and miRNA, was extracted from paraffin wax samples. Reverse transcription and RT-PCR method were used to detect the relative quantification of mi R-27a in both gastric cancer tissues and surrounding tissues. At the same time, the clinicopathological data of Tibetan patients with gastric cancer were collected and analyzed statistically. To search for the reports of miR-27a in gastric cancer tissues from other nationalities, and to make a comparative study. Results: (1) the expression level of MiR-27a was 7.415 鹵0.129 in Tibetan gastric cancer and 2.050 鹵0.062 in adjacent tissues of Tibetan cancer. The difference between the two groups was statistically significant (P0.05). That is, the expression of miR-27a in Qinghai Tibetan gastric cancer was higher than that in adjacent tissues.
