MiRNA-101在結(jié)直腸腺瘤及結(jié)直腸癌中表達(dá)差異的分析

【摘要】:結(jié)直腸癌是人類最常見的惡性腫瘤之一。其發(fā)病率在惡性腫瘤中分別占第三位,死亡率占惡性腫瘤的第四位,已經(jīng)嚴(yán)重威脅著人們的健康,并對(duì)患者家庭造成了很大的疾病負(fù)擔(dān)。部分患者在就診時(shí)已發(fā)生遠(yuǎn)處轉(zhuǎn)移,失去了手術(shù)機(jī)會(huì)。由于目前結(jié)直腸癌的發(fā)病率高,治療效果不佳,特別是晚期癌癥患者。因此,進(jìn)一步探討結(jié)直腸癌的發(fā)病機(jī)制,將為結(jié)直腸癌提供新的治療方向。與其他惡性腫瘤一樣,結(jié)直腸癌的發(fā)生一般是由癌基因的激活及抑癌基因的失活引起。但調(diào)節(jié)這些基因的生物學(xué)機(jī)制卻仍處于研究階段。自Lee等首次發(fā)現(xiàn)miRNA后,miRNA在生命活動(dòng)中的作用機(jī)制不斷被發(fā)掘,尤其是在腫瘤發(fā)生、發(fā)展中的作用。miRNA是一類高度保守的非編碼小RNA,控制著約三分之一的人類基因的表達(dá)。miRNA一般通過(guò)調(diào)控癌基因或抑癌基因的信號(hào)通路來(lái)促進(jìn)或抑制腫瘤的發(fā)生、發(fā)展,并在腫瘤的診斷、治療及預(yù)后方面也起著重要的作用。miRNA-101作為miRNA家族中的一類,在人類多種腫瘤中呈表達(dá)下調(diào),抑制腫瘤細(xì)胞的增殖,在腫瘤的發(fā)生、發(fā)展過(guò)程中扮演著抑癌基因的角色。既往有研究發(fā)現(xiàn)miRNA-101在結(jié)直腸癌中呈低表達(dá),并通過(guò)不同靶基因抑制結(jié)直腸癌細(xì)胞的增殖、轉(zhuǎn)移。但這些研究大部分是體外研究,且臨床樣本量較少。因此,本研究在這些研究的基礎(chǔ)上擴(kuò)大臨床樣本量,分析miRNA-101在結(jié)直腸腺瘤、結(jié)直腸癌組織中的表達(dá)情況,揭示其如何介導(dǎo)結(jié)直腸癌的發(fā)生,有助于為結(jié)直腸癌以后的診斷和治療提供新的途徑。目的:本研究將檢測(cè)miRNA-101在結(jié)直腸腺瘤及結(jié)直腸癌組織中的表達(dá)是否有差異,并探討其在結(jié)直腸癌發(fā)生過(guò)程中的作用。方法:本研究采用熒光定量PCR技術(shù)測(cè)定33例結(jié)直腸腺瘤患者(管狀絨毛腺瘤8例,管狀腺瘤25例)腺瘤組織及腺瘤旁正常組織,以及31例結(jié)直腸癌癌組織及癌旁正常組織中miRNA-101的相對(duì)表達(dá)量。本研究使用SPSS22.0統(tǒng)計(jì)學(xué)軟件分析數(shù)據(jù),因數(shù)據(jù)呈偏態(tài)分布,故采用配對(duì)設(shè)計(jì)符號(hào)秩和檢驗(yàn)和兩獨(dú)立樣本的秩和檢驗(yàn),P0.05認(rèn)為差異有統(tǒng)計(jì)學(xué)意義。比較miRNA-101在腺瘤及癌中的表達(dá)是否有差異,分析其表達(dá)量與年齡、性別、病理類型等各臨床病理資料之間的關(guān)系,并分析miRNA-101在結(jié)直腸癌的發(fā)生、發(fā)展中的作用。結(jié)果:(1)在結(jié)直腸腺瘤患者中,miRNA-101在腺瘤中的表達(dá)高于腺瘤旁組織(P0.05);(2)在腺瘤患者中,miRNA-101在不同性別,年齡及病理類型(即管狀絨毛狀腺瘤與管狀腺瘤相比)中的相對(duì)表達(dá)量無(wú)差異,無(wú)統(tǒng)計(jì)學(xué)意義;(3)在結(jié)直腸癌患者中,miRNA-101癌組織中的表達(dá)低于癌旁組織(P0.05);(4)miRNA-101在結(jié)直腸癌患者在患者的各臨床特征間(即性別、年齡、腫瘤部位)的相對(duì)表達(dá)量無(wú)差異,無(wú)統(tǒng)計(jì)學(xué)意義。結(jié)論:miRNA-101在腺瘤組織中表達(dá)升高,在結(jié)直腸癌組織中表達(dá)降低,提示其從正常結(jié)直腸組織到結(jié)直腸腺瘤,結(jié)直腸腺瘤到結(jié)直腸癌的發(fā)生發(fā)展過(guò)程中發(fā)揮了調(diào)控作用。本研究為miRNA-101作為結(jié)直腸癌預(yù)測(cè)、預(yù)后指標(biāo)提供了實(shí)驗(yàn)基礎(chǔ)。
[Abstract]:Colorectal cancer is one of the most common malignant tumors in humans. The incidence of the cancer is the third in the malignant tumor, and the death rate is the fourth of the malignant tumor, which has seriously threatened the health of the people, and has caused a great burden on the patient's family. Some of the patients had a distant metastasis at the time of the visit and the opportunity was lost. Because of the high incidence of colorectal cancer, the treatment effect is not good, especially in the patients with advanced cancer. Therefore, a further study of the pathogenesis of colorectal cancer will provide a new direction for the treatment of colorectal cancer. As with other malignant tumors, the occurrence of colorectal cancer is generally caused by the activation of the oncogene and the inactivation of the tumor suppressor gene. But the biological mechanism of regulating these genes is still in the research stage. After the first discovery of miRNAs, such as Lee, the role of miRNAs in life activities has been discovered, especially in the development of tumorigenesis and development. miRNA is a highly conserved non-coding small RNA that controls about one-third of the human gene expression. The miRNA is generally used to promote or inhibit the occurrence and development of the tumor by regulating the signal path of the oncogene or the tumor suppressor gene, and also plays an important role in the diagnosis, treatment and prognosis of the tumor. The miRNA-101 is used as a class in the miRNA family, the expression of the miRNA-101 is down-regulated in a plurality of tumors of the human, the proliferation of the tumor cells is inhibited, and the role of the tumor suppressor gene is played in the development and the development process of the tumor. Previous studies have found that miRNA-101 has a low expression in colorectal cancer and inhibits the proliferation and metastasis of colorectal cancer cells by different target genes. However, the majority of these studies were in vitro and the clinical sample size was small. Therefore, this study expanded the clinical sample size on the basis of these studies, and analyzed the expression of miRNA-101 in colorectal adenoma and colorectal cancer, and revealed how to mediate the occurrence of colorectal cancer and to provide a new way for the diagnosis and treatment of colorectal cancer. Objective: To study the expression of miRNA-101 in colorectal adenoma and colorectal cancer, and to explore the role of miRNA-101 in colorectal cancer. Methods: The relative expression of miRNA-101 in 33 patients with colorectal adenoma (8 cases of tubular villus adenoma, 25 cases of tubular adenoma), normal tissue of the adenoma, and 31 cases of colorectal cancer and the normal tissues of the adjacent normal tissues were determined by fluorescence quantitative PCR. In this study, the data was analyzed using the SPSS22. 0 statistical software, and the rank sum test of the paired design symbol and the rank and test of the two independent samples were used because of the bias distribution of the data, and the difference was considered to be of statistical significance. To compare the expression of miRNA-101 in adenoma and carcinoma, and to analyze the relationship between the expression of miRNA-101 and the clinical and pathological data of age, sex and pathological type, and to analyze the role of miRNA-101 in the development and development of colorectal cancer. Results: (1) In the patients with colorectal adenoma, the expression of miRNA-101 in the adenoma was higher than that of the adjacent adenoma (P0.05). (2) In the patients with adenoma, the relative expression of miRNA-101 in the different sex, age and pathological type (i.e., the tubular villous adenoma and the tubular adenoma) was not different. (3) The expression of miRNA-101 in the patients with colorectal cancer is lower than that of the adjacent tissues (P0.05). (4) The relative expression of miRNA-101 in the patients with colorectal cancer (i.e., sex, age, tumor site) has no difference and no statistical significance. Conclusion: The expression of miRNA-101 in the tissue of colorectal cancer is increased, and the expression of miRNA-101 in colorectal cancer is reduced, suggesting that it plays an important role in the development of colorectal adenoma, colorectal adenoma and colorectal cancer. The present study provided an experimental basis for miRNA-101 as a prognostic indicator for colorectal cancer.
