芒柄花黃素對人胃癌細(xì)胞株MKN-45增殖、凋亡的影響及其機(jī)制
[Abstract]:Objective to observe the effect of Flavatin on proliferation and apoptosis of human gastric cancer MKN-45 cell line and to explore its possible mechanism. Methods Human gastric cancer MKN-45 cells at logarithmic growth stage were divided into two groups: control group and control group. The control group was cultured with 20 ~ 4080 渭 g / m ~ (-1) amaranthin, and the control group was cultured on a complete medium without Acanthoxanthin. MTT assay was used to observe the proliferation of the cells in each group for 72 h. The effect of DAPI staining on the morphology of MKN-45 cells was evaluated. Phosphorylated I 魏 B (p-I 魏 B) and NF- 魏 B p65 were detected by Western blotting assay at 48 h after administration. Results the inhibition rate of cell proliferation was increased with the increase of the concentration of anthocyanin, and the inhibition rate of cell proliferation was increased with the increase of intervention time (P 0.05). At 72 h after administration, the apoptotic rates in the control group were 25.62% 鹵2.57% 鹵48.27% 鹵3.18%, respectively, and 72.551% 鹵4.3551% 鹵0.54% in the control group, respectively, compared with those in the control group (P 0.05). The relative expression of p-I 魏 B and NF- 魏 B p65 protein in the cells of Agni BU C group was higher than that in the control group at 48 h, and the relative expression of p-I 魏 B protein and NF- 魏 B p65 protein increased with the increase of Flavatin concentration (P 0.05). Conclusion Acanthopanthin can inhibit the proliferation of human gastric cancer MKN-45 cell line and promote apoptosis. The mechanism may be the activation of NF- 魏 B signaling pathway by Acanthopanthin.
