TRAF6表達與胃癌患者營養(yǎng)狀況及預(yù)后的關(guān)系

【摘要】:目的目前有關(guān)腫瘤壞死因子受體相關(guān)因子6(tumor necrosis receptor-associated factor 6,TRAF6)在胃癌相關(guān)領(lǐng)域的研究仍有限。本研究通過檢測胃癌患者組織中TRAF6表達,探討其與患者營養(yǎng)狀況和預(yù)后的關(guān)系,為胃癌治療提供新思路。方法采用逆轉(zhuǎn)錄-聚合酶鏈反應(yīng)、蛋白質(zhì)印跡法、酶聯(lián)免疫吸附試驗檢測2008-01-01-2013-06-30浙江省人民醫(yī)院102例胃癌患者及53例腹腔良性疾病患者組織(腹直肌、血清)中TRAF6mRNA和蛋白的表達情況,并結(jié)合臨床資料分析其對胃癌患者營養(yǎng)狀況及預(yù)后的影響。結(jié)果胃癌患者腹直肌中TRAF6 mRNA的相對表達量(3.89±0.09)較腹腔良性疾病患者(1.77±1.08)明顯升高,t=13.050,P0.001。蛋白質(zhì)印跡法結(jié)果顯示,胃癌患者腹直肌中TRAF6蛋白表達較腹腔良性疾病患者明顯升高,P0.05。胃癌患者血清中TRAF6蛋白的相對表達量(4.89±1.09)較腹腔良性疾病患者(2.78±1.18)明顯升高,t=14.595,P0.001。胃癌患者血清中TRAF6蛋白表達水平與患者體質(zhì)量丟失(χ~2=12.901,P0.001)、血清白蛋白(χ~2=17.422,P0.001)及血紅蛋白水平(χ~2=4.886,P=0.027)有關(guān)。胃癌患者血清中TRAF6蛋白表達水平與患者胃癌病理類型(χ~2=6.154,P=0.046)、淋巴結(jié)轉(zhuǎn)移(χ~2=10.237,P=0.001)及TNM分期(χ~2=7.727,P=0.005)有關(guān)。胃癌患者血清中TRAF6蛋白表達水平與患者生存率有關(guān),χ~2=6.641,P=0.010。結(jié)論胃癌患者腹直肌及血清中TRAF6mRNA和蛋白表達升高,且血清中TRAF6蛋白表達水平患者的營養(yǎng)狀況及預(yù)后存在關(guān)聯(lián)。TRAF6可能與胃癌患者癌性惡病質(zhì)的發(fā)生發(fā)展有關(guān)。
[Abstract]:Objective at present, the research on tumor necrosis factor receptor related factor 6 (tumor necrosis receptor-associated factor 6 TRAF 6 is still limited in the field of gastric cancer. The aim of this study was to investigate the relationship between TRAF6 expression and nutritional status and prognosis of patients with gastric cancer, and to provide a new approach for the treatment of gastric cancer. Methods 102 patients with gastric cancer and 53 patients with benign abdominal diseases (rectus abdominis) were detected by reverse transcription-polymerase chain reaction (RT-PCR), Western blot and enzyme linked immunosorbent assay (Elisa). The expression of TRAF6mRNA and protein in serum and its effect on nutritional status and prognosis of patients with gastric cancer were analyzed with clinical data. Results the relative expression of TRAF6 mRNA in rectus abdominis in gastric cancer patients (3.89 鹵0.09) was significantly higher than that in benign abdominal diseases (1.77 鹵1.08). The results of Western blotting showed that the expression of TRAF6 protein in rectus abdominis of gastric cancer patients was significantly higher than that in patients with benign abdominal diseases (P0.05. The relative expression of TRAF6 protein in serum of patients with gastric cancer (4.89 鹵1.09) was significantly higher than that of patients with benign abdominal diseases (2.78 鹵1.18). The expression of TRAF6 protein in serum of patients with gastric cancer was correlated with the loss of body mass (蠂 ~ 2 ~ 2 ~ (12.901) P ~ (0.001), serum albumin (蠂 ~ (2) ~ (2) ~ (17.422) P ~ (0.001) and hemoglobin (蠂 ~ (2) ~ (4.886) P ~ (0.027). The expression of TRAF6 protein in serum of patients with gastric cancer was correlated with the pathological type of gastric cancer (蠂 ~ 26.154), lymph node metastasis (蠂 ~ 2 + 10.237) and TNM stage (蠂 ~ 27.727 P ~ (0.005). The expression of TRAF6 protein in serum of patients with gastric cancer was correlated with survival rate (蠂 ~ 2 ~ 2 ~ (6.641). Conclusion the expression of TRAF6mRNA and protein in rectus abdominis and serum of gastric cancer patients is increased, and the nutritional status and prognosis of patients with serum TRAF6 protein expression are related. TRAF6 may be related to the occurrence and development of cancer cachexia in gastric cancer patients.
