【摘要】：[目的]探討PD-L1通過STAT3抑制血管內(nèi)皮細(xì)胞增殖遷移的影響。[方法]實(shí)驗(yàn)分組:A:pcDNA3.1+siRNA-Control(NC組);B:pcDNA3.1+siRNA-PD-L1(si-PD-L1組);C:STAT3+siRNA-Control(STAT3組);D:STAT3+siRNA-PD-L(STAT3+si-PD-L1組),按照分組做細(xì)胞轉(zhuǎn)染。通過MTT、Cell-LightTMEd U細(xì)胞增殖檢測轉(zhuǎn)錄因子STAT3、siRNA-PD-L1對(duì)血管內(nèi)皮細(xì)胞增殖率的影響;通過細(xì)胞劃痕實(shí)驗(yàn)檢測0、24、36 h融合率、Western Blot檢測轉(zhuǎn)錄因子STAT3、siRNA-PD-L1對(duì)血管內(nèi)皮細(xì)胞增殖標(biāo)志物PCNA表達(dá)水平的影響。[結(jié)果]C組HUVEC細(xì)胞的增殖率比A組顯著增高60%,共轉(zhuǎn)pcDNA3.1-STAT3+siRNA-PD-L1后D組細(xì)胞增殖率比C組顯著下降52%。劃痕實(shí)驗(yàn)劃痕融合率C組比A組顯著增高(p0.05),D組與C組相比降低(p0.01)。PCNA表達(dá)C組顯著高于其他組(p0.05),D組與C組相比顯著降低40%(p0.01)。[結(jié)論]STAT3顯著促進(jìn)HUVECs增殖遷移能力,而干擾PD-L1以后,可下調(diào)細(xì)胞增殖遷移。
[Abstract]:[objective] to investigate the effect of PD-L1 on proliferation and migration of vascular endothelial cells through STAT3. [methods] A:pcDNA3.1 siRNA-Control group (NC group); B:pcDNA3.1 siRNA-PD-L1 group (si-PD-L1 group); C:STAT3 siRNA-Control group (STAT3 group); D:STAT3 siRNA-PD-L group (STAT3 si-PD-L1 group). The effect of transcription factor STAT3,siRNA-PD-L1 on the proliferation of vascular endothelial cells was detected by MTT,Cell-LightTMEd U cell proliferation, and the effect of transcription factor STAT3,siRNA-PD-L1 on the expression of PCNA in vascular endothelial cells was detected by cell scratch assay. [results] the proliferation rate of HUVEC cells in group C was significantly higher than that in group A, and the proliferation rate in group D was significantly lower than that in group C after pcDNA3.1-STAT3 siRNA-PD-L1 transfer. The rate of scratch fusion in group C was significantly higher than that in group A (p0. 05), D group was significantly lower than that in group C (p0. 01). PCNA expression was significantly higher than that in other groups) (p0. 05), D group and group C significantly decreased by 40% (p0. 01). [conclusion] STAT3 can significantly promote the proliferation and migration of HUVECs, but after interfering with PD-L1, it can down-regulate the proliferation and migration of HUVECs.
