NOX通路在氧化應(yīng)激誘導(dǎo)的宮頸癌細(xì)胞凋亡中的作用

【摘要】:目的探討煙酰胺腺嘌呤二核苷酸磷酸氧化酶(NOX)通路在氧化應(yīng)激誘導(dǎo)的宮頸癌細(xì)胞凋亡中的作用。方法流式細(xì)胞術(shù)檢測(cè)不同類型宮頸癌細(xì)胞內(nèi)在的總活性氧(ROS)水平及NOX途徑抑制劑DPI處理前后不同類型宮頸癌細(xì)胞內(nèi)線粒體途徑和NOX途徑ROS水平;DPI預(yù)處理、H_2O_2作用后采用流式細(xì)胞術(shù)檢測(cè)不同類型宮頸癌細(xì)胞凋亡率。結(jié)果人乳頭瘤病毒(HPV)陽性宮頸癌細(xì)胞內(nèi)在ROS水平明顯高于HPV陰性宮頸癌細(xì)胞(P0.05),兩類宮頸癌細(xì)胞線粒體途徑來源ROS水平比較差異無統(tǒng)計(jì)學(xué)意義(P0.05)。與預(yù)處理前比較,DPI預(yù)處理后HPV陽性宮頸癌細(xì)胞內(nèi)在ROS水平均明顯降低(P均0.05)。與誘導(dǎo)前及HPV陰性宮頸癌細(xì)胞比較,H_2O_2誘導(dǎo)后HPV陽性宮頸癌細(xì)胞凋亡率均明顯升高,且高于DPI預(yù)處理及DPI預(yù)處理+H_2O_2誘導(dǎo)細(xì)胞(P均0.05)。結(jié)論 NOX通路在HPV感染導(dǎo)致宮頸癌的過程中具有重要作用,促氧化治療可能更好地殺傷HPV陽性宮頸癌細(xì)胞。
[Abstract]:Objective to investigate the role of nicotinamide adenine dinucleotide phosphate oxidase (NOX) pathway in oxidative stress induced apoptosis of cervical cancer cells. Methods flow cytometry was used to detect the intrinsic levels of total reactive oxygen species (ROS) in different types of cervical cancer cells and NOX pathway inhibitor (DPI) before and after treatment with different types of cervical cancer cells. The mitochondrial pathway and NOX pathway ROS level of different types of cervical cancer cells were pretreated with H _ 2O _ 2. Apoptosis rate of different types of cervical cancer cells was detected by flow cytometry. Results the ROS level of (HPV) positive cervical cancer cells was significantly higher than that of HPV negative cervical cancer cells (P0.05). There was no significant difference in ROS level between the two types of cervical cancer cells derived from mitochondrial pathway (P0.05). Compared with that before preconditioning, the ROS level of HPV positive cervical cancer cells was significantly lower than that of pretreated cells (P < 0. 05). The apoptosis rate of HPV positive cervical cancer cells was significantly higher than that of DPI pretreatment and DPI pretreatment (P < 0. 05). Conclusion NOX pathway plays an important role in the process of cervix cancer caused by HPV infection, and it may be better to kill HPV positive cervical cancer cells by the treatment of promoting oxidation.
