HDAC3與宮頸癌相關(guān)性研究

【摘要】:目的檢測宮頸癌組織中組蛋白去乙酰化酶3(HDAC3)的表達水平,分析其與宮頸癌臨床病理特征的關(guān)系。同時探討HDAC3小干擾RNA(si RNA)對宮頸癌Hela細胞株HDAC3的影響。方法采用半定量RT-PCR及免疫組化學(xué)方法(IHC)檢測12例正常宮頸組織、21例高級別上皮內(nèi)瘤變(CINⅡ~Ⅲ)組織和64例宮頸癌組織中HDAC3表達情況,分析其與宮頸癌臨床病理參數(shù)的關(guān)系;運用HDAC3的抑制劑恩替諾特(Entinostat,MS-275),以宮頸癌細胞Hela、Siha為細胞模型,檢測藥物對細胞活力、增殖、凋亡的影響。應(yīng)用si RNA干擾技術(shù)分別導(dǎo)入HDAC3干擾質(zhì)粒(HDAC3 si組)和空載對照質(zhì)粒(HDAC3Control組),采用半定量RT-PCR和Western bloting檢測干擾效果。結(jié)果HDAC3 m RNA在正常宮頸組織、高級別上皮內(nèi)瘤變及宮頸癌組織表達量分別為(2.125±0.537)、(1.386±0.793)、(1.0±0.212),差異有統(tǒng)計學(xué)意義(P0.05);HDAC3蛋白在正常宮頸組織、高級別上皮內(nèi)瘤變及宮頸癌組織陽性表達率分別為8%(1/12)、38%(8/21)、94%(60/64),差異有統(tǒng)計學(xué)意義(P0.05);免疫組織化學(xué)結(jié)果顯示癌組織中HDAC3表達強度與腫瘤分化程度、肌層浸潤及淋巴轉(zhuǎn)移有關(guān)(P0.05);HDAC3的選擇性抑制劑MS-275對Hela、Siha細胞具有抑制作用。與轉(zhuǎn)染空載對照質(zhì)粒相比,轉(zhuǎn)染HDAC3干擾質(zhì)粒后,宮頸癌Hela細胞株HDAC3 m RNA水平和蛋白表達量均明顯下降,比值分別為(0.923±0.028)vs(0.091±0.001)、(0.887±0.035)vs(0.093±0.005),差異有統(tǒng)計學(xué)意義(P0.05)。結(jié)論HDAC3可能參與了宮頸癌的發(fā)生、發(fā)展過程;HDAC3的選擇性抑制劑MS-275對Hela、Siha細胞具有抑制作用,這種作用可能通過抑制HDAC3實現(xiàn)。HDAC3 si RNA轉(zhuǎn)染宮頸癌Hela細胞株可以下調(diào)HDAC3 m RNA水平及蛋白的表達量,為宮頸癌的靶向治療提供一種新思路。
