載阿霉素介孔二氧化硅納米粒對人舌癌Tca-8113細胞增殖及凋亡的影響

【摘要】:目的:探討載阿霉素介孔二氧化硅納米粒(MSNs@DOX)對體外培養(yǎng)的人舌癌Tca-8113細胞增殖及凋亡的作用。方法:透射電鏡(TEM)和氮吸附法對介孔二氧化硅納米粒(MSNs)進行表征;紫外分光光度計檢測阿霉素(DOX)釋放行為;MTT法檢測細胞增殖抑制作用;Hochest33342染色、流式細胞術(shù)檢測細胞凋亡和周期分布;Western blot檢測P53、Bcl-2、Bax和Caspase-3蛋白表達。結(jié)果:MSNs形態(tài)規(guī)則,分布均一,平均粒徑、介孔孔徑、比表面積和孔容分別為約30nm,19.59682nm、193.4296m2/g、0.947625cm3/g;MSNs細胞相容性良好;與單獨DOX相比,載藥組的細胞增殖抑制率及凋亡率更顯著,且呈一定MSNs濃度依賴性(P0.05);細胞明顯阻滯于G0/G1期;P53、Bax、Caspase-3表達顯著升高,Bcl-2表達顯著降低。結(jié)論:該載藥納米傳輸系統(tǒng)有望用于舌癌治療。
[Abstract]:Aim: to investigate the effect of doxorubicin loaded mesoporous silica nanoparticles (MSNs@DOX) on proliferation and apoptosis of human tongue cancer Tca-8113 cells in vitro. Methods: the (MSNs) of mesoporous silica nanoparticles was characterized by transmission electron microscopy (TEM) and nitrogen adsorption, and the release behavior of doxorubicin (DOX) was detected by UV spectrophotometer. Apoptosis and cell cycle distribution were detected by flow cytometry. Western blot was used to detect the expression of Bcl-2 Bax and Caspase-3 protein. Results the morphology, distribution, mean particle size, mesoporous pore size, specific surface area and pore volume were about 30 nm ~ (-1), 19.59682 nm ~ (-1), 193.4296 m ~ (2 /) 路g ~ (-1) ~ 0. 47625 cm ~ (3) / g ~ (-1), respectively. Compared with DOX alone, the cell proliferation inhibition rate and apoptosis rate of drug loaded group were more significant. The expression of Caspase-3 was significantly increased in the G0/G1 phase and the expression of Bcl-2 was significantly decreased. Conclusion: the drug-loaded nano-delivery system is expected to be used in the treatment of tongue cancer.
