人外周血γδT細(xì)胞免疫治療裸鼠皮下人肝癌細(xì)胞移植瘤的體內(nèi)實驗研究
[Abstract]:Aim: to study the immunotherapy of human peripheral blood 緯 未 T cells in nude mice transplanted with human hepatoma cells (SMMC-7721). Methods: (1) Human hepatoma cell line SMMC-7721 was inoculated subcutaneously into BALB/c nude mice to establish hepatoma nude mice model. (2) Monocyte was extracted from healthy human peripheral blood and 緯 未 T cells were amplified in vitro. (3) the established nude mice were randomly divided into 5 groups. The positive control group was 5-fluorouracil (5-Fu), the negative control group was normal saline, the treatment group was injected into the tail vein of nude mice with different doses of 緯 未 T cells (1 脳 105 T5 脳 10 5 and 25 脳 10 5), and the positive control group was injected intraperitoneally with 5-Fu. The negative control group was injected with saline into tail vein of nude mice. The inhibitory effects of different doses of 緯 未 T cells on tumor were observed, including body weight, food intake and growth status before and after treatment. The changes of tumor volume (TV), relative to tumor volume (RTV) and tumor proliferation rate [T / C (%)] were compared with those of positive control group and negative control group. Results: different doses of 緯 未 T cells inhibited the growth of xenografts in nude mice to varying degrees (P0.05), and the growth of TV-positive cells was significantly lower than that of 5-Fu positive controls (P0.05). The inhibition degree of RTV in each dose group was similar, The relative tumor proliferation rate of each dose group was slightly lower than that of 5-Fu group, but there was no significant difference. Conclusion: human peripheral blood 緯 未 T cells have a significant inhibitory effect on hepatoma xenografts in nude mice and provide experimental evidence for the establishment of a new immunotherapy method for liver cancer.
